Effect Of Yiqi Huoxue Decoction On Angiogenesis And Expression Of VEGF And BFGF In The Ischemic Myocardium Of Rats

Objective: To investigate the effects of Yiqi Huoxue Decoction on the angiogenesis and the protein expression of vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(b FGF)in the ischemic myocardium of rats with acute myocardial infarction(AMI).Methods: The AMI model was established by ligating the left anterior descending coronary artery,Grouping by means of random digital tables: model group,sham operation group,Qishen Yiqi decoction group and Yiqi Huoxue decoction group.Determination of Endothelin-1(ET-1)in Serum by double Antibody Sandwich elisa,Determination of nitric oxide(NO)concentration by nitrate reductase.The microvessel count(MVC),microvessel density(MVD)and expression of VEGF and b FGF in the ischemic myocardium were detected by immunohistochemistry.The change levels of VEGF and b FGF protein in the ischemic myocardium were detected by Western blot.Results: The results of this study show,comparison with sham-operation,myocardial injury was observed in different degree in rats in each group,the level of ET-1 was significantly increased,NO level decreased significantly,increased MVC and MVD,increased expression of VEGF and b FGF proteins,the difference is significant(P <0.01).Compared with model group,the degree of myocardial tissue injury in Qishen Yiqi decoction group and Yiqi Huoxue decoction group was obviously alleviated compared with the former group,ET-1was significantly decreased,NO level significantly increased,MVC and MVD increased significantly,VEGF and b FGF protein expression increased significantly,the difference was also significant(P <0.01).Comparison with Qishen Yiqi decoction group,the degree of myocardial injury in the group of Yiqi Huoxue decoction was improved compared with the former group,ET-1level decrease,NO level rise,increased MVC and MVD,increased expression of VEGF and b FGF proteins,the difference is statistically significant(P <0.05).Conclusion: Yiqi Huoxue decoction group can promote myocardial angiogenesis after myocardial infarction in rats,on the one hand,it can reduce the concentration of ET-1 in serum and increase NO concentration,to protect the vascular endothelium.On the other hand,up-regulation of myocardial VEGF and b FGF protein expression may be one of the important mechanisms of action.
