Abstract/Summary:
BackgroundThe Bladder cancer is the most common malignant tumor in the urinary system of our country,bladder cancer patients are increasing at a rate of one hundred thousand annual in worldwide.The bladder cancer is the fourth most common malignancy in men,and it is the ninth place among women of the most common clinical malignancies.At present,resection of bladder tumor or radical cystectomy is the most important treatment for bladder cancer,however,early diagnosis is difficult,and although the method of surgery has made rapid progress,but the trauma of surgery can not be avoided,complications are greater.The 5 year survival rate is still low for the patients with advanced stage even they are treated with surgery.Carboxy-tone protein(CTEN)is one of the members of the family of TENSIN.Recent studies have found that CTEN has an obvious correlation with the invasiveness and prognosis of various tumors,it plays a key role in the occurrence and development of malignant tumors,and is an important prognostic factor.In this study,the expression of CTEN in bladder urothelial carcinoma tissues,normal pathological grades and clinical stages was detected by immunohistochemistry,The relationship between the expression level and the pathological indexes was also analyzed.In order to further investigate the role and clinical significance of CTEN in the carcinogenesis and development of bladder cancer,and provides a new approach for the clinical diagnosis,diagnosis,evaluation of prognosis,etiology and target therapy of bladder cancer.ObjectiveTo investigate the expression of CTEN in bladder urothelial carcinoma tissues and to compare with the healthy subjects at the same time,to analysis the clinical significance of CTEN in bladder urothelial carcinoma,and to provide theoretical basis for the evaluation of the disease.Methods82 cases of urothelial carcinoma of urinary bladder in Department of Urology,150th Central Hospital of PLA were collected in 2015~2016 years.All the patients underwent surgical treatment.The diseased tissue samples were taken and no radiotherapy or chemotherapy was performed before the operation.15 cases of the control group of were suspected bladder lesions,hospitalized patients,cystoscopy or lithotomy was performed during the hospitalization,and the bladder tissue was obtained at random during biopsy.Specimens of bladder urothelial carcinoma patients and controls were collected and immunohistochemical staining was performed.The staining results were observed and compared between groups.According to the depth of invasion,patients with urothelial carcinoma of bladder were divided into non-invasive invasive bladder cancer(below T2,also called superficial squamous cell carcinoma)and invasive bladder cancer(T2 and T2 or invasive bladder cancer).According to the degree of differentiation,patients are divided into high level and low grade.According to the age of the patients will be divided into<60 age group and above 60 years group.Patients were divided into male and female groups according to gender.The staining results were observed and compared between groups.Results1.The bladder urothelial carcinoma tissue is obviously stained,and the positive staining signals of CTEN protein were brownish yellow particles,located in the cell membrane and(or cytoplasm).The staining of normal bladder tissues is not obvious.CTEN was expressed in normal bladder tissues and bladder cancer tissues,The majority of normal bladder tissues were negative,with few positive expression,and the positive rate was 26.27%(4/15).The positive expression rate of CTEN was 63.41%(52/82)in BUC group,which was brownish yellow or dark brown.The difference between the two expressions was statistically significant(P<0.05).2.With the increase of pathological grade of bladder cancer,the expression rate of CTEN also increased,low grade papillary urothelial carcinoma the positive expression rate of 54.54%(30/55),high grade papillary iurothelial carcinoma the positive expression rate of 81.48%(22/27).The difference between the two groups was statistically significant(P<0.05).The expression rate of CTEN in Ta~T1 phase(superficial phenotype)was 54%(27/50),while the expression rate between T2 and T4 was 78.13%(25/32),and the difference between the two expressions was statistically significant(P<0.05).At the age of<60 patients,the expression rate of CTEN was 57.69%(15/26),and above 60 years old patients with bladder cancer.The expression rate was 66.07%(37/56),there was no significant difference between groups(P>0.05).The positive rate of CTEN in male patients with bladder cancer was 65.31%(32/49),while the positive rate of female patients was 60.61%(20/33),and there was no significant difference between groups(P>0.05)..Conclusions1.The expression of CTEN in bladder urothelial carcinoma tissues was significantly higher than that in normal bladder tissues.2.The expression level of CTEN in high-grade and high grade bladder urothelial carcinoma tissues is higher than that of low grade and low stage cancer.3.There were no significant differences in the expression of CTEN in the patients with urothelial carcinoma of the bladder between different sexes and ages.
