| Objective: Previous studies of the experimental group confirmed that acupuncture and “Neiguan” pretreatment can protect the heart from heart injury.We observed the effects of electroacupuncture and moxibustion “Neiguan” pretreatment on myocardial autophagy and apoptosis after myocardial ischemia / reperfusion injury in rats.Methods: SD rats were randomly divided into five groups: sham operation group,model group,ischemic preconditioning(IP)group,EA group and moxibustion group(n=8).Electroacupuncture group and moxibustion group were electroacupuncture or Moxibustion “Neiguan” 7 d before modeling,1 times a day,20 min each time.After seventh D pretreatment,coronary artery ligation was performed to establish the model of myocardial ischemia(ischemia 40 min)and myocardial ischemia-reperfusion injury(ischemia 40 min,reperfusion 1 h).HE staining and transmission electron microscopy were used to detect the pathological changes of left ventricular myocardium.The apoptosis and apoptosis index of cardiomyocytes were detected by Tunel.The expression of LC 3 and Beclin 1 protein in cardiac myocytes was detected by Western blot.Results:1.Effect of electroacupuncture and moxibustion Neiguan preconditioning on myocardial apoptosis index and autophagy related protein LC 3 injury in rats with myocardial ischemia.Compared with sham operation group,model group,apoptosis index was more higher(P < 0.05);compared with the model group,IP group,EA group,moxibustion group,apoptosis index were show a downward trend(P < 0.05);compared with IP group,the electroacupuncture group decreased the apoptotic index(P < 0.05),no significant difference between the moxibustion group(P > 0.05);compared with the EA group,moxibustion group,the apoptosis index was more higher(P < 0.05);compared with sham operation group,the expression of LC 3-II and the ratio of LC3-II/I of model group was significantly increased(P < 0.05);compared with themodel group,the expression of LC 3-II and the ratio of LC3-II /I values of IP group,EA group and moxibustion groupwere lower(P < 0.05);compared with IP group,the expression of LC 3-II and the ratio of LC 3-II/I of EA group was increased(P <0.05),but there the moxibustion group was no obvious difference(P > 0.05).2.Effects of electroacupuncture and moxibustion preconditioning on myocardial apoptosis index and autophagy related protein LC 3 and Beclin 1 in myocardial ischemia reperfusion rats The apoptotic index of the model group was significantly higher compared with the sham operation group(P <0.05).And the apoptotic index of the IP group,EA group and moxibustion group was significantly lower than the model group(P <0.05),and the apoptotic index in the EA group was lower than the IP group and the moxibustion group(P <0.05).Compared with the sham group,the ratio of LC 3-II/I and the expression of LC 3-II and Beclin 1 protein was significantly elevated(P <0.05);compared with the model group,the ratio of LC 3-II/I and the expression of LC 3-II and Beclin 1 protein of the IP group,EA group and moxibustion group was significantly decreased(P <0.05);Compared with the IP group,the ratio of LC 3-II/I and the expression of LC 3-II and Beclin 1 protein of the EA group was lower(P<0.05),but there the moxibustion group was no obvious difference.(P >0.05);Compared with the EA group,the ratio of LC 3-II/I and the expression of LC3-II and Beclin 1 protein of the moxibustion group was more higher(P < 0.05).Conclusion:Acupuncture and moxibustion preconditioning “Neiguan”can achieve similar protective effects of ischemic preconditioning and ischemia,in moderately enhanced myocardial cell autophagy,reduce cell apoptosis,improving cellular injury of myocardial ischemia caused by over expression during reperfusion;inhibition of autophagy in myocardial cell,prevent myocardial autophagy caused by excessive myocardial injury. |
