Objective:To study the chemical constituents from the Phellinus igniarius(L.ex Fr.)Quel and their anti-inflammatory activity.Methods:5 kg fruiting bodies of P.igniarius were extracted with 70%EtOH,and the crude EtOH extract was partitioned sequentially with CH2Cl2,EtOAc and n-BuOH.The EtOAc and n-BuOH extracts were purified by using normal and reversed silica gel,sephadex gel column chromatography and high performance liquid chromatography,respectively.The structures of the isolated compounds were elucidated by optical rotation,NMR and so on.The compounds were assayed for their inhibitory effects on the pro-inflammatory mediators nitric oxide(NO),tumour necrosis factor-a(TNF-a),and interleukin-6(IL-6)in lipopolysaccharide(LPS)stimulated mouse macrophages(RAW264.7)by Griess and ELISA methods.Results:Twenty compounds were isolated from the EtOAc extract and n-BuOH extract of P.igwniarius.The structures of compounds 1-20 were identified by comparing their physicochemical properties and spectral data with those of published in literatures as cerevisterol(1),?-sitosterol(2),betulinic acid(3),3?,6?,16?-trihydroxylup-20(29)-ene(4),monogynol A(5),acetylursolic acid(6),eburicol(7),ethyl ?-D-glucopyranoside(8),ethyl a-D-Galactoside(9),?-D-fructopyranose and ?-D-fructofuranose mixture(10),a-nigerose(11),trehalose(12),2-deoxy-D-ribono-1,4-lactone(13),(+)-taxifolin(14),cedrusin(15),ligstroside(16),oleuropein(17),amyl phthalate(18),2-ethylhexyl phthalate(19),1-0-(?-D-glucopyranosyl)-(2S,,3R,4E,8E)-2-[(2)-2-hydroxynonadecanoylamino]-9-methyl-4,8-octadecadiene-1,3-diol.Among them,compounds 4,9,16 and 17 were isolated for the first time from P.igniarius.The experimental results of anti-inflammatory activity showed that compounds 1,4,9-12,16,17,19 and 20 could significantly inhibit the production of NO,IL-6,and TNF-a of RAW264.7 cells induced by LPS at the concentrations of 3 ?M,10?M and 30?M,and reduced the inflammatory response.
