Phellinus igniarius (L.ex.Fr) Quel is well known in China as a medicinal fungus in folk medicine, which can date from Compendium of Materia Medica. However, this fungus is just investigated in China at present though being explored extensively in Europe and American, Korea and Japan. Due to its rarity in the nature and difficulty in culture, P. igniarius is mainly produced by submerged culture. In this paper, the comparative analysis of chemical composition from dry matter of culture broth (DMCB) from P. igniarius in submerged culture (PISC) and fruiting bodies of P. igniarius, the evaluation of anti-hepatoma effect in vivo and its possible active mechanism of DMCB and its fractions from PISC, optimization of extraction technique of polysaccharides from DMCB of P. igniarius by Response Surface Method (RSM) were studied.It's shown that the DMCB of P. igniarius in submerged culture was rich of crude protein, amino acid and polysaccharides, the contents of which were 34.28%, 19.64% and 24.90%, respectively. And unsaturated fatty acids were prior to saturated fatty acids, in which oleic acid and linoleic acid were the main components. There were also many other nutrition components in it. It's as well as the fruiting bodies of P. igniarius in many ways in nutrition and development value even then better.Both of DMCB and its fractions from PISC showed anticancer effect. The potency of DMCB and its fraction II (polysaccharides) seems to be better than the other two fractions, the inhibitory rate at the high dose (1000 mg/kg/d) of DMCB and fraction II was 33.5%, 40.3% respectively. Both of DMCB and fraction II could affect immunity index but below the level of natural group mice. The histopathological results suggested necrosis in H22 cells were induced in both of DMCB and fraction II groups, as proved by lower bcl-2 genes expression, higer bax genes expression. Both of DMCB and fraction of DMCB from PISC also increased SOD,GSH-Px enzymatic activity.The primary researchs shows polysaccharides fraction show the most significant antitumor effect in both of DMCB and its three fractions from PISC.The results showed that the optimum extraction rate by response surface analysis was up to 24.90% at 84℃for 2.7 h while ratio of water to material was 33.5∶1 (v/w). The polysaccharides extracted under optimum condition were analyzed by gas chromatography and infrared. The monosaccharide component of the polysaccharides was mannose 3.65%, glucose 12.00% and galactose 0.24%. Moreover, there were rich in glucoses in the products and the glycosidic bonds could beα-types mainly.
