Objective: Obesity is severely threat to human health.Acupuncture is an effective method to lose weight.Previous studies in our team found that acupuncture therapy played an active role in regulating the disorganized intestinal flora and energy metabolism.Therefore,it is essential to explore the effect and mechanism of acupuncture on intestinal flora and to provide theory for clinical acupuncture treatment of obesity.Methods: In this study,21 SD rats as the experimental object,were randomly divided into normal group with normal diet and control group with high fat diet according to the random sampling principle.The rats in the model group successfully became the nutritious obese rats during 7 weeks.And then they were randomly divided into treatment group and model group.The obese rats in the treatment group were treated the acupuncture therapy and chosen the acupoints: Zhongwan,Tianshu,GuanYuan,Fenglong,YinLingQuan,Zusanli.While the model group did not be taken any treatment.The treatment group and the model group continued to be fed with high fat diet and measured the rats' body weight,the length of tail and Lee 's index regularly at treatment period.After 4 weeks of treatment,the samples of rat blood lipid specimens and fecal specimens were successfully collected from the three groups.The rat blood lipid specimens and fecal specimens were determineed by ultraviolet spectrophotometry.The information of intestinal flora in the three groups were detected by 16 S rDNA technology.Analyzed the related datas by 16 S rDNA technology.Performed the statistical analysis by SPSS 17.Results:(1)Body weight and Lee 's index of rats induced by High fat diet was significantly higher than the normal group(P<0.01).(2)After acupuncture treatment,the body weight and Lee's index of rats in treatment group was lower than the model group(P<0.01).(3)The blood lipid of rats induced by high fat diet was significantly increased(P<0.01).And after treatment,the blood lipid of rats in treatment group was significantly decreased(P<0.01).(4)The intestinal flora of rats in treatment group was significantly different from that of rats in the model group.It was found that the proportion of Bacteroides,Firmicutes,Enterobacter and Enterococcus in treatment group was bigger than that of the model group.Conclusions: Obese rats induced by high fat diet might have dyslipidemia and intestinal flora disorder.Acupuncture therapy could significantly reduce the body weight and the Lee's index of obese rat.What's more,Acupuncture could regulate the disorder intestinal microflora of obese rat,improving the differences,abundance and distribution of intestinal microflora.We concluded that Firmicutes phylum including Enterococcus and Lactobacillus,Bacteroidetes phylum including Bacilli and Prytobacterium,Bifidobacterium in the actinomycetes phylum,Helicobacter and Escherichia,were related to loose weight by acupuncture therapy.
