Objective: Using the Sophora japonica cv.Jinhuai as the raw material,the extraction technology of rutin and quercetin was optimized to obtain the gist for the use of this medicinal material.Methods: Selecting rutin content as the dependent variable,on the basis of single factor test,the material-liquid ratio,methanol concentration and ultrasonic power were selected as independent variables.The ultrasonic-assisted extraction technology for Golden Sophora japonica rutin was optimized by response surface methodology,and performing prediction analysis.The content of rutin was mensurated by HPLC.(High Performance Liquid Chromatography): mobile phase methanol(A)-0.5% phosphoric acid water(B)gradient elution(0 to 12 min,10% to 75% A,12 to 24 min,75% to 88% A,24 ~35 min,88%-95% A),detecting the wavelength is 260 nm;quercetin content was selected as the dependent variable,on the basis of single factor test,ultrasonic time,material-to-liquid ratio,ultrasonic power were used as independent variables.The response surface method was used to optimize ultrasonic-assisted extraction technology for quercetin,and performing predictive analysis.The content of quercetin was mensurated by HPLC.The mobile phase methanol(A)-0.5% phosphoric acid water(B)gradient elution(0 to 12 min,10% to 75% A,12 to 24 min,75% to 88% A,24 ~ 35 min,88% ~ 95% A),detecting the wavelength was 360 nm;Results: The best extraction process conditions of rutin: the ultrasonic power was 250 W;methanol concentration was 90%;ultrasonic time was 60 min;material-liquid ratio was 1:50(g/mL).The predicted values and verification values of rutin in this extraction were 35.32 % and 33.19 % respectively,which were litter difference between the two.The optimal extraction process conditions of quercetin: ultrasonic time was 60 min,the ratio of material and liquid was 1:10,and the ultrasonic power was 350 W.Under this extraction condition,the predicted values of extraction efficiency of quercetin was 9.62%.the verification value was 9.97%,which was close to the predicted value.Conclusions: The optimal extraction process was stable and feasible with good predictability.It can improve the rational application of golden sophora japonica and provide reference for the preparations and development of rutin and quercetin in Sophora japonica.
