| Rutin is an important secondary metabolite in drying flower and bud from Sophara japonica ,the native tree of China. As the representative a flavonoids, Rutin has many physiological activities. It has the vitamin P effect, which maintains the resistance of the blood vessel, reduces the exceptional osmoses and brittleness of blood vessel, and reduces blood pressure, etc. It has been used in clinical treatment for the coronary dilating activity, adjuvant therapy for hypertension and arteriosclerosis. Therefore, it seem to be particularly important for factory that how to extract Rutin from material, theoretical study on basic structure and Pharmacological effect.Based on the present situation of traditional extraction technology of Rutin from Sophara japonica, this article comprehensively elaborates condition of the Ultrasonic Wave extraction. By single-factor experiment, then carries on the statistics analysis with the SAS software, selects appropriate experimental design. It is deduced that an optimal extracting process has been determined: room temperature, 20KHZ, 1000w, 1:15 (W/V), 10minutes, distill twice. In this situation, the extraction yield of Rutin is 19.16%.According to the literature report, The Rutin extraction from Sophara japonica which used Ultrasonic Assistance Technology is purified by the molecule morphology modification method,The Rutin with refined extracts is determined the purity by HPLC. The optimum technological condition of purification was obtained: normal temperature, putting solution of methyl alcohol [45~55%(v/v)]into crude Rutin extraction from Sophara japonica [Rutin 40%~50%(m/m)], then dissolving and refining two times. In this situation, Content of Rutinin in purified product is 90.36%.According to the literature and the author's research. The structure of chromium(Cr3+)complex with the Rutin from Sophara japonica are identified by AAS, UV, IR. The results show that 5-hydroxyl group,4-carbonyl group and Ortho-dihydroxyphenyl in B ring take part in the coordination with metal ions, The molar ratio of Rutin extraction from Sophara japonica and chromium (Cr3+)is 1:2. |
