Objective: Mastoidectomy obliteration is important for reconstruction of the ear ofthe anatomical structure, function and improve patient`s prognosis. But there are some inthe choice of packing materials, there are some possible disadvantages associated with theuse of Autogenous bone or artificial substitutes. The experiment was to explore whetherbone marrow-derived mesenchymal stem cells mixed with demineralized bone matrix canpromote osteogenesis in a mastoid obliteration procedure. Methods: Four male guineapig were used for obtaining the third generation of bone marrow mesenchymal stem cells(BMSCs), using Brdu-labeled BMSCs. Twenty guinea pig were equally divided into twogroups:1) the experimental group: implantation of xenograft demineralized bone matrixcombined with bone marrow stromal cells in guinea pig’s tympanic bulla;2) the controlgroup: implantation of Autogenous bone in guinea pig’s tympanic bulla. computedtomography were obtained from each bulla at1and2months postsurgery. the amount ofbone were blinded evaluation with scale, the two group were compared through observingthe amount of bone, rank sum test was used for analysis. Guinea pig were killed atpostoperative moths two for routine histology. Results: The mortality of two groups wasno difference in the end of experimental. The experimental group was a amount of boneformation at the2month postoperative, as compared to the control group (P<0.05).Implantation of xenograft demineralized bone matrix combined with bone marrowmesenchymal stem cells in tympanic bulla can well promote osteogenesis in a mastoidobliteration procedure. Conclusion: The successful formation of bone using xenograftdemineralized bone matrix combined with bone marrow mesenchymal stem cells to repaircritically sized bulla defect in the guinea pig model was accompanied with the Autogenous bone graft.
