Objective:The family of aquaporins discovered in the early 1990s,have been t he focus of research in recent years.Its discovery marks a new milesto ne in understanding how water molecules work in cells.Aquaporin 2(A QP2)is one of the most pathophysiolo-gically significant aquaporins found so far,and is an essential substance to maintain the water balan ce of the body.Diuretics are widely used in clinic,especially in the treatment of co ngestive heart failure.Although some of the current studies relate to th e effects of diuretics on urine AQP2,but little is known about changes in AQP2 genes and proteins in the kidneys.In this experiment,the rat model of water load was established.The expression of AQP2 and AQP2mRNA in urine and kidney was taken as an index.To observe the effects of Furosemide?Acetazolamide.low d ose of Wuling Powder?normal dose?high dose on the metabolism of water solution in rats with water load.Method:Healthy SD rats,male,weighing about 200g,provided by Experimen tal Animal Center of Heilongjiang University of traditional Chinese Me dicine.Feeding Rats in Animal Laboratory of Heilongjiang University of tra ditional Chinese Medicine(barrier environment),feed and water on ti me every day,adapt to a week.Screening of urinary function in rats bef ore the experiment,Rat fasting can not help but water 12hours,drench physiological saline of 25ml kg-l,rats with more than 40%of the amo unt of urine collected within 2 hours can be used for diuretic action.Selection of 60 qualified rats,divided randomly into 6 groups,Normal group?Furosemide group?Acetazolamide group?Wuling Powder Low Dose Group?Dose group in Wuling Powder?Wuling Powder High Dos e Group.After the water load test is over,put rats in metabolic cages i mmediately(One rat per cage).Before the experiment,the rats fasted and could not help the water.On the second day,the rats in each grou p were given intragastric administration according to the volume of 20 ml·kg-1,the urine volume was collected and measured at 1h,2h,4h aft er gavage,respectively.Continuous gastric perfusion for 10 days,collect 24 hours urine volume on day 10.After the experiment,the rats were killed immediately.Intraperitoneal anesthesia with pentobarbital sodium in rats,after collecting blood,the rats were killed and their kidneys were rem oved quickly.A kidney is put into formaldehyde for use in immunohist ochemical experiments,and the other is wrapped with tin foil paper an d put into liquid nitrogen to be frozen,then put it in the cryogenic r efrigerator at-80 ? for Western Blot and RT-PCR experiment.Result:Compared with the control group,the urine volume in the furosemid e group,acetazolamide group and Wuling powder equivalent dose grou p was significantly increased(P<0.05).There was no significant diffe rence in Wuling Powder low dose group and Wuling Powder high dose group compared with the control group(P>0.05).Compared with the control group,the renal AQP2 expression in the furosemide group,acetazolamide group and Wuling powder equivalent dose group was significantly lower than that in the control group(P<0.05).There was no significant difference in renal AQP2 between Wuli ng Powder low dose group and Wuling Powder high dose group(P>0.05).Compared with the control group,the expression of AQP2mRNA in t he kidney of furosemide group,acetazolamide group and Wuling powde r group was significantly lower than that of control group(P<0.05).There was no significant difference in renal AQP2mRNA expression be tween Wuling Powder low dose group and Wuling Powder high dose g roup(P>0.05).Conclusion:1.In this experiment,the difference between traditional diuretic W uling Powder and western medicine diuretic was observed by establishi ng water loaded rat model.The results showed that Wuling Powder ha d obvious diuretic effect and had long term effect.2.The equivalent dose group(0.26 g/kg)of furosemide,ac etazolamide and Wuling Powder had better diuretic effect on water-1 oaded rats,and AQP2 and AQP2 mRNA expression in kidney tissues of rats were reduced.The diuretic effect of Wuling Powder in low dose group(0.13 g/kg)and high dose group(0.52 g/kg)was not obvious.
