Objective:By observing the effect of Wuling powder on the expression of renal aquaporin2 gene in rats with kidney-yang deficiency edema model,the mechanism of the effect of Wuling powder on kidney-water metabolism was investigated.Methods:60 healthy SPF SD rats were divided into blank group(10),the remaining 50 rats by intraperitoneal injection of rats hydrocortisone induced joint in the rat tail intravenous injection of adriamycin solution in the rat model with kidney Yang deficiency edema,were randomly divided into model group,control group and drug group(WLS-L,WLS-M and WLS-H dose group),each group of 10;The urine volume at each stage of the experiment was recorded.The pathological changes of renal tissue were observed by HE staining.The serum levels of c AMP,c GMP,T3,T4,E2 and T were determined by enzyme-linked immunosorbent assay.IHC was used to determine the distribution and expression of AQP2 in renal tissue.The expression of AQP2 m RNA in renal tissue was determined by Q-RT-PCR.The expression of AQP2 protein in kidney was detected by WB.Results:1.After the model was replicated,the signs of the rats were consistent with those of kidney-yang deficiency syndrome in traditional Chinese medicine;In addition,the urine output of the model group was reduced at 24 h,and the urine protein excretion of the model group was increased at different degrees at 24 h.Moreover,the serum levels of c AMP,c GMP,c AMP/c GMP,T4,T and E2/T in the model group were significantly different from those in the blank group,indicating that the hormone levels of the endocrine system were all disrupted,suggesting that the model was successfully replicated.2.After administration,the rats in the WLS-M and WLS-H dose groupsreturned to normal defecation,freely moved,and their physical signs improved to different degrees.Compared with the model group,there was no significant difference between the WLS-L group and the F group,while there was a significant difference between the WLS-M and WLS-H dose groups(P<0.01).Compared with the model group,the WLS-M and WLS-H dose groups all showed stable urine volume increase on day 1 or 7 and 14 after administration,with significant difference(P<0.01).Compared with the model group,the serum levels of c AMP,c GMP,c AMP/c GMP,T4,T and E2/T in the WLS-M and WLS-H dose groups were significantly different(P<0.01).After HE staining,the renal tubules and interstitium were improved in the WLS-M and WLS-H dose groups.After immunohistochemical staining,compared with the model group,the difference in AQP2 expression in the WLS-L group was only statistically significant(P<0.05),and there were significant differences in AQP2 expression in the other groups(P<0.01).No matter at the m RNA level or protein level,AQP2 in the drug administration group was significantly lower than that in the model group,and the difference was statistically significant(P<0.01).Conclusion:Studies have shown that wuling powder may have a significant effect on the treatment of edema,and the potential mechanism at least involves hormone regulation.By improving the endocrine state of rats with kidney-yang deficiency edema,the general state of rats with kidney-yang deficiency edema can be improved,and the pathological damage of the kidney can be alleviated.At the same time,wuling powder can down-regulate the expression of AQP2 in kidney,inhibit the reabsorption of water,and improve the edema state of rats with kidnia-yang deficiency edema.Therefore,wuling powder has a therapeutic effect on kidnia-yang deficiency edema,and its molecular mechanism for treating edema may be related to the down-regulation of AQP2 expression in kidney.
