Effect Of Buyanghuanwu Decoction On The Expression Of Reg-2,mTOR,Olig-2 In Rats With Spinal Cord Injury

Objective: To research the influence of Buyanghuanwu decoction to the motor function of spinal cord injury rats and express of regenerating gene protein-2,the mammalian target of rapamycin and oligodendrocyte transcription factor-2,use Methylprednisolone as a contrast,and then investigating the mechanism of action of Buyang Huanwu decoction to intervene spinal cord injury.Methods: 24 rats were divided into 4 groups randomly with CONTROL group,SCI model group,BYHWD model group and MP model group(6 each).Using methods of Allen's to establishing SCI model,and at the time of 1 day after,3 days after,5 days,7 days after when the model set up successfully chosen Basso Beattie Bresnahan Iocomotor rating scale and Inclined plane test to evaluate the rats' restoration effect.CONTROL group had no special treatment except perfused with the same volume of saline for 7 days.When spinal cord injured,SCI model group perfused with the same volume of saline for 7 days.When spinal cord injured,BYHWD model group administrated Buyang Huanwu decoction by drenching for 7 days.When spinal cord injured,MP model group gave peritoneal injection methylprednisolone for 7 days.The rats were sacrificed at 7 days which had a sufficient depth of anesthesia before after operation,then remove injured spinal segment about one centimeter with HE staining,and use immunohistochemical,immunofluorescence and Western blot to detect the expression of Reg-2?mTOR?Olig-2 in rats spinal cord tissue.Results: 1.Basso Beattie Bresnahan Iocomotor rating scale and Inclined plane test in SCI model group have significantly decreased;for the first day and three days after being given the drug,Basso Beattie Bresnahan Iocomotor rating scale and Inclined plane test in BYHWD model group and MP model group haven't statistically significant(P>0.05).For the five days after being given the drug,Basso Beattie Bresnahan Iocomotor rating scale and Inclined plane test in BYHWD model group and MP model group have better restoration effect than SCI model group,have statistically significant(P<0.05),BYHWD model group and MP model group haven't statistically significant(P>0.05).For the seven days after being given the drug,Basso Beattie Bresnahan Iocomotor rating scale and Inclined plane test in BYHWD model group and MP model group have much better restoration effect than SCI model group,have statistically significant(P<0.01),BYHWD model group and MP model group haven't statistically significant(P>0.05).2.The result of HE staining shown that injured spinal segment tissue became irregularly arranged,scar formation and part of nerve cell structure obscured,inflammatory cell infiltration.For the seven days after being given the drug,spinal cord tissue orientation of fibrils in BYHWD model group and MP model group had definite directionality,neurogliocyte multiplication and inflammatory cell infiltration decreased.3.Compared with CONTROL group,the expression of Reg-2 and mTOR lies in spinal cord tissue in SCI model group have significantly increased(P<0.05).For the seven days after being given the drug,the expression of Reg-2 and Olig-2 lies in spinal cord tissue in BYHWD model group and MP model group have significantly increased(P<0.05),the expression of mTOR have significantly decreased(P<0.05).Compared with MP model group,the expression of mTOR in BYHWD model group have significantly decreased(P<0.05).Conclusions: 1.Buyang Huanwu decoction can facilitate the restoration of injured spinal cord rats,its effect the same as methylprednisolone.2.Buyang Huanwu decoction can reduce injured spinal cord rats neurons apoptosis,promote the growth of neurogliocyte,inhibits inflammatory cell infiltration.3.Buyang Huanwu decoction can elevate the expression of Reg-2 and Olig-2,restrain the expression of mTOR.4.The treatment of Buyang Huanwu decoction to spinal cord injury may relevant to elevate the expression of Reg-2 and Olig-2,restrain the expression of mTOR,facilitate the division growth activity of neurogliocyte,attenuate the inflammatory reaction,and then play the function of protect neuronal.
