| Spinal cord injury is a serious disease of the central nervous system.With the development of social economy,the incidence of spinal cord injury increases grad-ually.Spinal cord injury has the characteristics of high disability rate and high mortality,and the pathological process after spinal cord injury is complicated.This paper mainly explores the effect of Buyang Huanwu decoction on NLRP1(NOD-like receptor protein 1)/Caspase-1(cysteinyl aspartate specific proteinase-1)pathway of pyrop-tosis after spinal cord injury.This paper includes two parts:literature review and experimental study.The first part:literature reviewThis part includes two literature reviews.The first literature review introduced the role of NLRP1 inflammasome in traumatic central nervous system injury.This ar-ticle mainly reviews from four sections:the structure of NLRP1 inflammasome,the expression of NLRP1 inflammasome induced by traumatic central nervous system injury,the activation and regulation of NLRP1 inflammasome in traumatic central nervous system injury and NLRP1 inflammasome as a target therapy after trau-matic central nervous system injury.The second part expounds the research pro-gress of Buyang Huanwu Decoction in spinal cord injury,and summarizes the pathological mechanism of spinal cord injury,the pharmacological study of Buyang Huanwu Decoction and the role of Buyang Huanwu Decoction in spinal cord injury.Part two:experimental study Objective:To investigate the effect of Buyang Huanwu Decoction on NLRP1/Caspase-1 pathway of pyroptosis in rats with acute spinal cord injury.Methods:32 SPF rats were randomly divided into four groups:sham operation group,methylprednisolone group,Buyang Huanwu Decoction group and model group.After the model was established successfully,the decoction of Buyang Huanwu Decoction con-taining granule 2g/mL was intragastrically infused with 1.75ml/kg body weight once a day for 14 days in Buyang Huanwu Decoction group.Methylprednisolone group was injected with methylprednisolone via caudal vein within 24 hours after successful modeling,the first dose was 30mg/kg,the rest was 5.4mg/kg/h,once every 4 hours,and the sham operation group and model group were intragastrically infused with normal saline of 1.75ml/kg body weight once a day for 14 days.The neurological recovery of the rats was evaluated by BBB scale on the first,third,seventh and fourteenth days after intervention.The recovery of nerve function was evaluated by BBB scale at 1,3,7,14 days after treatment.The protein expression of nlrpl,Caspase-1 and IL-18 in SCI tissue was detected by Western Blot method.Results:BBB scale:There was no change in the scale of rats in the sham operation group.The scale of Buyang Huanwu Decoction group and methylprednisolone group was higher than that of the model group 3 days,7 days and 14 days after treatment,(P<0.05).At 7d and 14d after treatment,,the scale of the rats in each group did not change significantly.The protein expression of nlrp1,Caspase-1 and IL-18 in in SCI tissue of rats on the 14th day after interven-tion:the protein expression of nlrp1,Caspase-1 and IL-18 in Buyang Huanwu Decoction group and methylprednisolone group was lower than those in sham operation group and higher than those in model group(P<0.05).There was no significant difference between Buyang Huanwu Decoction group and methylprednisolone group.(P>0.05).Conclusion:The therapeutic effect of Buyang Huanwu Decoction combined with spinal cord decompression on acute spinal cord injury is obvious,which can effec-tively interfere with spinal cord injury and promote the recovery of neurological function after acute spinal cord injury in rats.Its mechanism may be to reduce pyroptosis death by inhibiting NLRP1/Caspase-1 pathway.However,the specific mechanism of Buyang Huanwu Decoction action in NLRP1/Caspase-1 pathway of pyroptosis needs to be further studied. |
