Objective: 1)To explore the associations of genetic polymorphisms of FBN1 and TGF-?1 gene with Debakey type III aortic dissection and its risk factors in Chinese Han population in Xinjiang;2)to investigate the relationship between FBN1 and TGF-?1 gene mutation and prognosis of Debakey type III aortic dissection.Methods: A case-control study including 159 cases with Debakey type III aortic dissection confirmed by aortic CTA and 241 cases gender and age negative coronary angiography controls was desighed for the present study.Using qRT-PCR methods,to detect FBN1,TGF-? 1 gene rs145233125,rs201170905 label SNP and rs1800469 genotyping,and the three SNP were tested in the distribution of AD group and control group,and the analysis of genotype and allele.T test,logistic regression analysis,COX Regression Analysis were and Kaplan-Meier survival curves used to explore the relationship between genotypes of different loci and genetic risk and risk factors of Debakey III aortic AD.Results:Rs145233125,rs201170905 and rs1107064 in the FBN1 gene and rsrs180469 in theTGF-?1 gene was associated with Debakey type III aortic dissection in Han ethnic in Xinjiang.Rs201170905 GG,rs1107064 GG and rs180469 TT genotype significantly increased the risk of Debakey type III aortic dissection,after adjustment for other factors,rs201170905,rs1107064 and rs180469 gene polymorphism was still independent risk factors for Debakey type III aortic dissection,Kaplan-Meier survival curves showed that the additive model of rs201170905 of FBN1 gene was related to the survival of Debakey III AD.Similarly,we were also found that rs1800469 of TGF-?1 gene was related to the survival of AAD in the additive model.Using COX Regression Analysis,after adjusting for other confounding factors,we found no significantly associated between rs145233125,rs201170905,rs11070646 and rs1800469 and the prognosis of Debakey III AD.Conclusion: 1)Genetic polymorphisms of FBN1 and TGF-?1 were associated with Debakey type III aortic dissection in Han ethnic,which maybe result from alteration of plasma GLU,triglyceride and and inflammatory cells with genetic mutation in FBN1 and TGF-?1;2)FBN1 and TGF-?1 gene polymorphism were associated with clinical prognosis and survival rate in Debakey type III aortic dissection.
