Research background:At present,cancer has become one of the most important factors threatening human healthy and life,according to the survey,In China the incidence and mortality of cancer increased year by year in the past 10 years,breast cancer is still the most common tumor in women's cancer.tumor incidence rate is high and rising about 3-4%a year.It is expect that new cancer cases in that United States will reach 878 980 in women in 2018,226120 cases of them are breast cancer,accounting for about 30%of the National Women's expected cancer incidence.The 5 year survival rate of breast cancer patients can reach 90%after receiving scientific and standardized comprehensive treatment in the early stage of disease.Due to some patients still lack of consciousness or accept inappropriate treatment of breast cancer,breast cancer progress to late stage.The high heterogeneity of mammary tumor cells makes traditional treatments unable to meet the individualized treatment needs of breast cancer patients.Molecular biology technology has been widely applied in the research of the occurrence,development,metastasis and drug resistance mechanism of breast cancer,making the diagnosis and treatment mode of breast cancer to change from evidence-based medicine to precision medicine.Precise medical mode has brought new choice for patients with advanced breast cancer.Under the traditional treatment mode,there are many problems:improper treatment leads to the recurrence,metastasis of breast cancer,excessive treatment leads to unnecessary pain for the patients,some molecular subtypes of breast cancer lack of treatment and have poor prognosis,exploring resistant mechanism of breast cancer,founding the new key oncogene,exploring new tumor therapeutic targets and applying to clinical therapy is particularly important.Objective:To observe the expression of PRR11 protein in breast cancer tissues,paraneoplastic tissues,benign breast tumors and normal breast tissues,analyze the relationship between PRR11 protein expression and clinicopathological parameters of breast cancer,and explore the effect of PRR11 protein on the prognosis of breast cancer.Methods:Selected 97specimens from patients who admit Department of Breast Surgery in the Southwest Medical University Affiliated Hospital from January 2011 to January2012,The patients are diagnosed with Non-special type invasive carcinoma of breast and their clinical data were improved.Collecting the paraffin embedding tissue of the tumor and of 3-5cm adjacent to malignant tumor.Selected 50specimens from patients who diagnosed with benign breast tumor,collecting the paraffin embedding tissue of the benign tumor and 0.5cm adjacent to the benign tumor.Use the immunohistochemical Envision method to detect the expression of PRR11 protein in all tissue specimens.Then use chi-square test to analyze the diffrence among the expression of PRR11 in 4 groups and the relationship between expression of PRR11 and the clinicopathological parameters of breast cancer.Log-rank test was used to compare the survival of PRR11 positive expression and negative expression group in breast cancer group.Use the Cox proportional hazard model to analyze if the expression of PRR11 is reviewed as a prognostic indicator in patients with breast cancer survival.Results:1.The expression level and expression rate of PRR11 in breast cancer tissue were significantly higher than those in adjacent tissue,benign tumor tissue and normal breast tissue.The difference was statistically significant(X~2=89.529,P<0.001).The other three groups had no significant difference in the positive rate of expression.2.We analysised the relationship between the expression level of PRR11 protein and clinical pathological parameters in the chi square test,we found that the expression level of PRR11protein is related to tumor size,pathological stage(TNM),histological grade,distant metastasis,hormone receptor status,Ki-67 expression(P<0.05).The expression of PRR11 increased with the tumor pathological staging(TNM),Ki-67 index and histological grade.3.The progression-free survival of breast cancer patients with positive expression of PRR11 is shorter.The progression-free survival of PRR11 positive expression was 51 months,and the negative expression was 77 months.(P<0.001).Tumor size,tumor pathological staging(TNM)and PRR11 expression were the prognostic indicators of breast cancer patients(P<0.05).Conclusion:1.The expression of PRR11 was negative or low In normal breast tissues,slightly up-regulated in benign tumor tissues and adjacent tissues,and increased significantly in breast cancer tissues.2.the expression level of PRR11 protein is related to tumor size,pathological stage(TNM),histological grade,distant metastasis,hormone receptor status,Ki-67 expression.The expression of PRR11 protein in breast cancer tissues with larger tumor diameter,distant metastasis,late stage and negative hormone receptor is relatively high.3.The expression of PRR11 was strongly correlated with tumor malignancy,and its expression increased with the rising of tumor stage,Ki-67 index and histological grade(decreasing of cell differentiation degree).4.The progression-free survival of breast cancer patients with positive expression of PRR11 is shorter.Tumor size,pathological stage(TNM)and PRR11 expression status are prognostic indicators of breast cancer.PRR11 may have guiding value for the diagnosis of breast cancer cell malignancy.
