Object:By observing the expression of cell membrane molecules and NF-kappa B expression in mouse uterine decidua antigen,the effect and mechanism of sympathetic nerve on the local immune microenvironment of mice uterus were investigated.Method:The 40 Kunming female mice that were randomly divided into two groups:20 of these were in the control group(group A,saline injection group)and the otherss were in experimental group(group B,6-hydroxydopamine injection).The mice of group A were injected 0.2ml of sterile saline(dissolved in sterile saline containing ascorbic acid in 0.01%)by intraperitoneal injection at 2:00 pm everyday.The mice of Another group were injected 0.2ml(0.01%ascorbic acid solution containing 6-hydroxydopamine solution)daily by intraperitoneal injection at 2:00 pm,after five days of continuous injection to respectively according to the ratio of 2:1 were caged.The first day of pregnancy is denoted as D1,then turn to cut out the uterus of mice at 14:00 of D1,D3,D5,D7,D9,Immunohistochemistry method was used to detect the expression of CD23,CD11b and CD209 in the uterus of mice.The mRNA expression of CD23,CD209 and CD11b was detected by in situ hybridization,and the content of NF-kappa B in uterus of mice was detected by EILSA.Results:(1)Compared with the control group,the level of mRNA and protein expression of CD23,CD11b and CD209 in uterine tissue of experimental group were lower than that in D1,D3 and D9,and there is a significant difference.In all parts of the uterus,the expression of CD23,CD11b and CD209 in the endometrium was the most.All the indexes were reduced at all time points except D5 that were injected6-hydroxydopamine.(2)In the control group,the protein and mRNA expression of CD23,CD11b,CD209were the most at D3,and then began to decline with time and gradually became stable,the experimental group reached the peak in D5 and then began to decline.(3)The content of NF-kappa B in the uterus in the experimental group was increesaed and up to a significant level after the injection of 6-hydroxydopamine,but it began to decline on D5.Conclusion:(1)Sympathetic nerve may regulate CD23~+B cells,CD209~+dendritic cells and CD11b~+macrophages to form a favorable maternal fetal immune tolerance environment,these three types of antigen presenting cells are similar to the direction of change trend.(2)By regulating the expression of CD23,CD209 and CD11b in decidual antigen presenting cells,it may be one of the ways that sympathetic nerves participate in the formation of local immune tolerance.(3)Sympathetic nerve may regulate the expression of CD23,CD209 and CD11b through NF-kappa B signaling pathway to regulate the function of antigen presenting cells.
