Cervical cancer threatening women's health seriously,because th e incidence of it is high as well as high mortality.The incidence of cervical cancer in our country ranks second in the world which is six times that of developed countries and is tending to younger.There are about 150,000 new cases of cervical cancer in China each year,accounting for about one-third of the total number of patients worldw ide.Compared with squamous cell carcinoma,adeno carcinoma having a high Incidence rate and high mortality that increasing gradually.The treatment of chemotherapy and surgery-based is attention as are sult that cervical adenocarcinoma is poor sensitivity to radiotherapy,chemotherapy is receiving increasing attention for Patient's high deman ds on quality of life after treatment and the desire for normal endocr ine function and sexual life.Paclitaxel is the first-line chemotherapy drug for cervical cancer,it has so significant effect on cervical cance r chemotherapy that it is widely used in cervical cancer patients in v arious stages.Paclitaxel is target to microtubules,by inhibitting their depolymerisation,Paclitaxel can arrest tumor cells in the G2/M-phase of the cell cycle.Adverse drug reaction and drug resistance of paclit axel are still limitting the clinical efficacy.Hence,exploring new anti-tumorred drugs to enhance sensitivity to existing drug resistance che motherapy has become a hot research incervical adenocarcinoma treat ment.Tetramethylpyrazine(TMP)is known as 2,3,5,6-Tetramethylpyra zine,a active component isolated from the Chinese herb Rhizoma Chuan qiong,with the functions suchas vasodilator,lower blood press ure,anti-myocardial,brain and other organ ischemia,blood stasis and so on.We used CCK-8 staining detect the influence of Ligustrazine ang Taxol alone or incombination on the cervical adenocarcinoma Hela cell;Annexin V-PI double staining detect the influence of two drugs alone or incombination on the Hela cell;SP-immunohstochemistry measure the effect of expression of PD-L1 in Hela cell when Ligustrazine ang Taxol alone or incombination on it.We can know that not only Paclitaxel but also Ligustrazine alone can inhibit the proliferation of cervical cancer Hela cells.When Ligustrazine and Taxol alone or incombination on the Hela cell,we can see Hela cells changing ther morphological with microscope.The change of Paclitaxel is superior than Ligustrazine;the morphological variety of Paclitaxel and Ligustrazine combination group is more than Ligustrazineor Taxol alone group,and the difference was statistically significant(P<0.05).The Annexin V-PI double staining showed that paclitaxel and tetramethylpyrazine could promote the apoptosis of cervical cancer Hela cells when they were used alone,and the effect of the paclitaxel was better than the ligustrazine;The apoptosis of the cells in Ligustrazine combine with Paclitaxel group was significantly higher than that of the first two groups alone,and the difference was statistically significant(P<0.05).SP immunohistochemistry results revealed that the expression of PD-L1 on the surface of Hela cells in paclitaxel group was slightly higher than that in control group;PD-L1 expression on Hela cells in Ligustrazine group was lower than that in paclitaxel group;The expression level of PD-L1 on the surface of the Ligustrazine combine with Paclitaxel group was significantly lower than that of the former two groups alone,and the difference was statistically significant(P<0.05).To sum up,we think paclitaxel and Ligustrazine alone can inhibit proliferation and promote apoptosis of cervical cancer Hela cells,Suggesting that botjh paclitaxel and Ligustrazine may have anti-cervical cancer effects,and the effects of paclitaxel are superior to those of Ligustrazine.The combined effects of paclitaxel and ligustrazine are significantly better than that of two drugs alone,we think that they have a synergistic effect on cervical cancer,the latter can enhance the efficacy of paclitaxel in cervical cancer chemotherapy.The antitumor mechanism of paclitaxel and ligustrazine treating on cervical cancer Hela cells may be related to the expression of PD-L1 on the cell surface.
