Part 1 Effect of sex, age, axial length and intraocular pressure on retinal layer thickness in normal eyes measured by spectral-domain optical coherence tomographyObjective:To quantitatively observe the effect of sex, age, axial length and intraocular pressure on the retinal thickness of normal subjects by spectral domain optical coherence tomography(SD-OCT).Methods:sixty-eight healthy volunteers included in this study. The male-to-female ratio is 28:40,the age range is from 25 to 80 years, and the axial length ranged from 22.0 to 25.0mm,intraocular pressure range is from 9?20mmHg.The average thickness retinal nerve fiber layer(RNFL)and total layers of retina(R),inner retinal layer(IRL)and retinal outer layer, namely photoreceptor layer(PL)were measured by SD-OCT with automated stratified technique respectively. The circle of the macular area with fovea as the center point, three concentric circles divided the region of macular with diamenters of 1mm,3mm and 6mm respectively(including the partition of R,R-1,R-3,R-6,IRL-1,IRL-3,IRL-6,PL-1,PL-3 and PL-6).Correlation analysis was conducted to analyze the correlation between those 10 parameters of structure thickness and sex, age, axial length or intraocular pressure respectively. Results:The R,R-1,R-3,R-6,IRL-1,IRL-3,PL-3 and PL-6 of female in the normal subjects were significantly thinner than those of the male(all P<0.05).Control the effect of sex,the thickness of RNFL around the optic disc was significantly thinning(all P<0.05),and most especially, the R,R-3,R-6,IRL-3 and IRL6 of macular area are significantly thinning(all P<0.05)with age.The factors of axial length and intraocular pressure have no correlation with the structure thickness of RNFL in the optic disc and macular area. Conclusion:In the normal population, the retinal thickness in the macular area of the female is significantly thinner than those of the male. With age,the retinal thickness in the macular area becomes significantly thinner.Part 2 Spectral domain optical coherence tomography quantifies the retina thickness in patients with type 2 diabetes mellitus without significant diabetic retinopathyObjective:To observe the retinal thickness of macular in type 2 diabetic mellitus(T2DM)patients without significant clinical features of diabetic retinopathy(DR).Methods:40 patients(40 eyes)in T2DM without DR and 68 healthy volunteers(68 eyes)were enrolled in this study.Using spectral domain optical coherence tomography(SD-OCT)and the software of automatic segmentation to measure the average thickness of total layers of retinal(R),inner retinal layer(IRL)and outer retinal layer namely photoreceptor layer(PL)in macular. In the foveal center, three concentric with the diameter is lmm,3mm and 6mm in the round(including the partition can be divided into R,R-1,R-3,R-6,IRL-1,IRL-3,IRL-6,PL-1,PL-3 and PL-6),while compare the average retinal thickness of retina between these two groups. Results:The mean thickness of PL-1,PL-3 and PL-6 in no DR group was significantly thinner than those in normal group[(71 ±3)?M vs.(73±3)?m;(66±2)?m vs.(67±2)?m;(64±2)?m vs.(64±2)?m,respectively](all P<0.05).In the male, the mean thickness of PL-3 in no DR group was significantly thinner than those in normal group[(67 ± 2)?m vs.(68 ± 2)?m](P<0.05).In the female, the mean thickness of PL-1,PL-3 and PL-6 in no DR group was significant thinner than those in normal group[(71 ± 4)?m vs.(73 ± 3)?m;(65 ± 2)?m vs.(67 ± 2)?m;(63 ±2)?m vs.(64± 2)?m,respectively](all P<0.05).There are no obviously differences in the other parts between these two groups. Conclusion:The mean retinal thickness of the parafovea and perifovea were significantly thinner in no DR group than the normal group. The measurement of the PL thickness of macular by SD-OCT may promote the study of early stage of DR.
