Objective:To observe the effects of miRNA-25 expression and tyrosinase level in the lesion area by using 308 excimer laser combined with CO2 lattice laser to intervene the vitiligo guine a pig model,and to analyze the possible correlation between o f them.Then explore the efficacy and possible mechanisms of vitiligo.Methods:43 healthy black guinea pigs?clean grade?were rand omly divided into healthy group?control group?and model gro up;8 of them were healthy group and 35 of them were vitilig o model preparation group.In the vitiligo model preparation gr oup,the guinea pigs were selected to make an area of approxi-mately 4cm×4cm in size on the back to be regularly depilated,and 3%hydrogen peroxide solution was applied externally twi ce a day?morning and evening?,until the back skin became lighter or whiter for 70 days.A randomized vitiligo guinea pig model was used for pathological biopsy.After confirming the success of the model,two vitiligo models were randomly select ed for the determination of the minimum erythema dose and th e selection of the CO2 lattice laser energy.After making vitilig o model successfully,32 guinea pigs were randomly divided in to 4 groups:the model group,the 308 excimer light group,the CO2 lattice laser group,and the combination group;plus 8healthy guinea pigs as the healthy group,a total of 5 groups.All groups except for the healthy group were intervened for 6weeks.In the end,HE staining and macroscopic evaluation of pigmentation were performed.The expression of miRNA-25 in lesions was determined by qRT-PCR and the tyrosinase conten t in the lesions was determined by ELISA.Results1.Compared with the miRNA-25 expression level in the health y group?0.95±0.13?,the expression level of mi RNA-25 in the model group?4.00±1.36?was significantly higher?p<0.05?;afte r 6 weeks of intervention,Compared with the model group,th e expression levels of miRNA-25 in the 308 excimer light gro up?2.89±0.01?,the CO2 lattice laser group?3.24±0.43?,and the combined group?1.20±0.15?were significantly decreased,and the difference was statistically significant?p<0.05?;Compared with the 308 excimer light group and the CO2 lattice laser gro up,the expression of miRNA-25 in the combination group was more significantly decreased?p<0.05?.2.Compared with the healthy group's tyrosinase content?48.81±2.06ng/L?,the tyrosinase content in the model group was sign ificantly decreased?20.22±0.61ng/L?,and the difference was sta tistically significant?p<0.05?;treatment 6w After that,compare d with the model group,308 excimer light group tyrosinase co ntent?36.27±4.10ng/L?,CO2 lattice laser group tyrosinase conte nt?30.30±3.11ng/L?and combined group tyrosine The content of acidase?42.11±2.71ng/L?was significantly increased?p<0.05?.The tyrosinase content in the combined group was significantl y higher than that of the 308 excimer light group and the CO2 lattice laser group.Statistical significance?p<0.05?.Conclusion:Single-use 308 excimer laser,CO2 laser and the combination o f both can promote skin lesion resurfacing in the vitiligo guine a pig model.The combination of the two is more significant a nd may be related to increasing local tyrosinase content and re ducing the expression of mi RNA-25 in lesions.
