Background:In recent years,the incidence of infertility has increased year by year.About 15%of couples of child-bearing age in the world are not pregnant for 1 years and need to be treated with intervention,of which the male factor is 50%.Endogenous antioxidants have some advantages in the treatment of oligoasthenospermia due to their good safety,less adverse reactions and definite curative effect.Lipoic acid(LA)is a natural powerful antioxidant.Recent studies have shown that LA can be used for the treatment of male infertility.LA can improve the semen quality of the patients with idiopathic asthenospermia and enhance the antioxidant capacity of the semen.LA has a good prospect in the treatment of male infertility associated with oligoasthenospermia.Object:To study the lipoic acid improved the reproductive function through regulating energy metabolism and oxidative stress in rat with oligoasthenospermia induced by omidazole(ORN).Methods:70 SD rats,with a weight of 230±20 g,were randomly divided into 7 groups,with 10 rats in each group.Group A:1 ml CMC-Na+10.5%ml olive oil;group B:400 mg/kg ORN+1 ml olive oil;group C:400 mg/kg ORN+50 mg/kg LA;group D:400 mg/kg ORN+100 mg/kg LA;group E:800 mg/kg ORN+1 ml olive oil;group F:800 mg/kg ORN+50 mg/kg LA;group G:800 mg/kg ORN+100 mg/kg LA.The rats were given the drug for 20 days,and the rats were weighed.The rats were anesthetized by intraperitoneal injection.The sperm concentration and vitality were detected by the tail of the epididymis,and the other epididymis and testis were observed by histomorphology.70 SD rats,with a weight of(230±20)g.Rats are treated with the same steps as above.The blood of the rat heart was taken and the serum levels of T,LH,FSH and INHB were detected.Rat testis and epididymis were taken to detect testicular ACP,AKP,LDH and SDH activities,epididymal alpha-glucosidase,LC content,OCTN2 protein expression,testicular and epididymal SOD,CAT,GSH-Px activity,MDA content,TAC and Nrf2 protein expression level.Results:1.Compared with the group A,the weight gain,testis and epididymal index were significantly decreased in group E(P<0.01).Compared with group E,the weight gain,testis and epididymal index were significantly increased in group F and group G(P<0.05 or P<0.01),and there was no significant difference between seminal vesicle index between groups.2.Compared with group A,sperm motility were significantly decreased in group B(P<0.01),and sperm count and motility were significantly decreased in group E(P<0.01),but there was no significant difference in sperm count among rats in groups B,C and D.Compared with group B,the sperm count and motility were significantly decreased in group E(P<0.01),and the sperm motility was significantly increased in group C and D(P<0.01).Compared with group E,the sperm count and motility were significantly increased in group F and G(P<0.05 or P<0.01).Compared with group F,the sperm motility was significantly increased in group G(P<0.01)3.There was no obvious change in the histomorphology of testis and epididymis in group A,B,C and D.The damage changes of the testis and epididymis were observed in group E.Compared with the group E,the histomorphology of the testis and epididymis was significantly improved in group F and G.4.Compared with group A,the serum level of INHB was significantly decreased in group B(P<0.05).The serum T,INHB were significantly decrease while FSH and LH were significantly increased in group E(P<0.01).Compared with group E,the serum FSH was significantly decreased in group F(P<0.01),T,FSH,LH and INBH were significantly improved in G group(P<0.05 or P<0.01).Compared with group F,the serum FSH,LH level was significantly decreased in group G(P<0.05).5.Compared with group A,the activity of ACP and SDH in testis was significantly decreased in group B(P<0.05),and the activity of ACP,AKP,LDH and SDH in testis was significantly decreased in group E(P<0.01).Compared with group E,the activity of ACP was significantly increased in group F(P<0.01).The activity of ACP,AKP,LDH and SDH in the testis was obviously improved in group G(P<0.05 or P<0.01).6.Compared with the A group,the alpha-glucosidase and LC content of the epididymis and the expression level of OCTN2 protein were significantly decreased in group B and E(P<0.05 or P<0.01).Compared with the group B,the content of alpha-glucosidase in the epididymis of was significantly increased in group C(P<0.05),and the indexes were significantly,increased in group D(P<0.01).Compared with group E,the epididymal alpha-glucosidase,LC content and expression level of OCTN2 protein were significantly improved in groups F and G(P<0.05 or P<0.01).7.Compared with group A,the activities of SOD and GSH-Px in testicular tissue were significantly decreased in group B(P<0.05).Compared with group A,the oxidative-antioxidant indexes were significantly different in epididymis of group B and in testis and epididymis of groups E(P<0.05 or P<0.01).Compared with group B,the activity of SOD and GSH-Px and TAC in epididymis increased significantly in group C(P<0.05 or P<0.01),and the testis SOD,epididymal SOD,GSH-Px activity,TAC and the expression level of Nrf2 protein was significantly increased in D group(P<0.05 or P<0.01).Compared with group E,the GSH-Px activity,TAC and expression of Nrf2 protein were significantly increased in group F(P<0.05 or P<0.01),MDA content was significantly decreased(P<0.05),and the oxidant-antioxidant indexes of testis were significantly improved in group G(P<0.01).The activity of SOD and CAT in epididymis were significantly increased in group F(P<0.05),MDA content was significantly decreased(P<0.01),and the oxidative-oxidative indexes of epididymis were significantly improved in group G(P<0.05 or P<0.01).Conclusion:LA can improve the reproductive through regulating energy metabolism and oxidative stress in rat with oligoasthenospermia induced by ORN.It can be used as a good choice for the treatment of oligoasthenospermia.
