Objective To study Effect of the petroleum ether extract from Amygdalus mongolica on blood lipid level,liver function and lipid peroxidation in hyperlipidemia rats,screen the dose-effect relation of hyperlipemia,analysis the components of the petroleum ether extract from Amygdalus mongolica and investigate the mechanisms of hyperlipemia;To investigate the hyperlipemia effect,the components,security and mechanism of Amygdalus mongolica oil.Method(1)Model was established by feeding high-fat five week,Experimental animal were divided into six groups:control group,model group,and 1.5g/kg,2.0g/kg,2.5g/kg of the petroleum ether extract from Amygdalus mongolica,Hyperlipidemic indicators and liver function were tested after collecting serum samples.After the rats were sacrificed,analyze the effect of antioxidative index.(2)Model was established by feeding high-fat five week,Experimental animal were divided into eight groups:control group,model group,and0.1875g/kg,0.375g/kg,0.75g/kg,1.5g/kg,3.0g/kg of the petroleum ether extract from Amygdalus mongolica,the method of detect indicator is same to fist one.the fatty acid composition of the petroleum ether extract from Amygdalus mongolica was analyzed by GC-MS.(3)Model was established by feeding high-fat five week,Experimental animal were divided into six groups:control group,model group,and 0.75g/kg,1.25g/kg,1.75g/kg of the petroleum ether extract from Amygdalus mongolica,the method of feeding and preparing model are same to second one,Blood and tissue were collected to detect Hyperlipidemic and antioxidative indicators.By examined the tissue histopathology.(4)Based on the single factor experiment,the effects of ratio of material to liquid,ultrasonic time and extraction temperature on the yield of Amygdalus mongolica Oil were studied through orthogonal experiment.(5)The components of Amygdalus mongolica oil was analyzed by GC-MS;Evaluate security by investigated for the acute toxicity.(6)Model was established by feeding high-fat five week,Experimental animal were divided into eight groups:control group,10ml/kg of control group,model group,and2.5ml/kg,5.0ml/kg,7.5ml/kg,10ml/kg of Amygdalus mongolica oil,the method of feeding and preparing model are same to fist part,Blood and tissue were collected to detect Hyperlipidemic and antioxidative indicators.By examined the tissue histopathology.Results(1)Compared with the hyperlipidemic model group,1.5g/kg of petroleum ether extract group from Amygdalus mongolica could significantly decreased the level of serum TC?LDL-C in the hyperlipidemia rat and TC/HDL-C?LDL-C/HDL-C?AST?AI,while MDA were lower(p<0.01,p<0.05).The activity of serum GSH-Px and GSH were higher(p<0.01,p<0.05);Medium dose group increased the activity of serum SOD?POD?CAT?GSH-px and GSH(p<0.01,p<0.05);High dose group increased the activity of serum POD and GSH(p<0.05).(2)0.75g/kg and 1.5g/kg of petroleum ether extract group from Amygdalus mongolica could significantly decreased the level of serum TC,LDL-C and MDA,The activity of serum SOD and GSH were higher;0.375g/kg of dose group increased the activity of serum SOD;Hyperlipidemic indicators of rat were change by petroleum ether extract group from Amygdalus mongolica in the dose-effect relationship and the dose rang is 0.5-1.5g/kg;the unsaturated fatty acid of Amygdalus mongolica Oil was up to 73.20%,among which oleic acid content is 28.87%,linoleic acid content is 38.68%,and the oil also contained some saturated fatty acids,which was hexadecanoic acid(19.66%).(3)1.25g/kg,1.75g/kg of petroleum ether extract group from Amygdalus mongolica could decreased the level of serum TC and All groups decreased the level of serum LDL-C after administered with drug for 4 weeks.Administered with drug for 7 weeks,petroleum ether extract group from Amygdalus mongolica could significantly decreased the level of serum TC,LDL-C,MDA and MDA from tissue,low does group was decreased TC/HDL-C,LDL-C/HDL-C and AI,which increased the level of serum SOD,GSH.Medium dose group decreased LDL-C/HDL-C,which increased the activity of serum SOD,GSH-PX and SOD from tissue.High dose group increased the activity of serum GSH,GSH-PX and GSH from tissue.Slighter pathological changes were observed in the liver of petroleum ether extract group from Amygdalus mongolica treated rats,Moreover,the low-dose group and the control group had similar pathological changes.(4)The optimum technology parameters were:extracting with the material-liquid ratio of 1:9 at 75°C for 3h and two times,under these conditions,the extraction ratio of Amygdalus mongolica Oil was 87.51%.(5)The GC-MS result showed the compositions ofthe Amygdalus mongolica oil were particularly rich in unsaturated fatty acids(76.55%),which were linoleic acid(47.11%),and octadecenoic acid(23.81%),and the oil also contained some saturated fatty acids,which was hexadecanoic acid(21.73%);It showed that the rats were secure by feeding the seed of oil at the dose of 21.5g/kg after14 days.(6)Without 2.5ml/kg of GSH-PX,SOD/MDA and 5.0ml/kg of SOD/MDA.Amygdalus mongolica Oil decreased the level of serum TC,LDL-C,MDA and TC/HDL-C,LDL-C/HDL-C,AI,which increased the activity of SOD,GSH,GSH-PX.Amygdalus mongolica Oil decreased hepatic index and MDA from tissue,10ml/kg and 2.5ml/kg dose groups were increased the activity of SOD,GSH from tissue,Slighter pathological changes were observed in the liver of petroleum ether extract group from Amygdalus mongolica oil-treated rats,Moreover,the 7.5ml/kg dose group and the olive oil group had similar pathological changes.Conclusion Petroleum ether extract group from Amygdalus mongolica has blood lipid-lowering,In a certain range,content with the reduction of dosing dose of cholesterol-lowering effect better,play a role of lipid may be associated with rich in unsaturated fatty acids,and the mechanism may be related with liver lipid metabolism improvement and antioxidant function enhancement.Amygdalus mongolica oil has rich in unsaturated fatty acids,blood lipid-lowering and fatty liver-preventing effect,dose and effect have a certain concentration-response relationship,the mechanism may be related with liver lipid metabolism improvement and antioxidant function enhancement.
