Objective: To analyze the preparation of silk fibroin/graphene oxide composite membrane composite materials and the effect of silk fibroin/graphene oxide composite membrane in treatment of long bone nonunion.Methods: To preparation fibroin solution and graphene solution.Take fibroin solution and graphene solution synthesis into silk fibroin/graphene oxide composite film.Take scanning electron microscopy to observe the morphological structure.Compound culture silk fibroin/graphene oxide composite membrane with 0.1wt%,0.2wt%,0.5wt%,1.0wt% graphene oxide and mouse embryonic osteoblast MC3T3-E1.Test the proliferation level of mouse embryonic osteoblast MC3T3-E1 by MTT colorimetry on 1d,3d,5d,7d.Make tibial defect nonunion model by 30 New Zealand white rabbit.30 cases New Zealand white rabbit were randomly divided into observation group and control group,each of 15 cases.Observation group was treated by silk fibroin/graphene oxide composite film composite and autogenous bone graft treatment.Control group was treated by autogenous bone graft treatment.Evaluated the bone defect repair by general,histological,biomechanical testing and X-ray on 4 weeks,8 weeks and 12 weeks after treatment.Results: The proliferation rate of MC3T3-E1 cells gradually decreased with the increase of graphene oxide concentration,the difference was statistic significant(P<0.05).The proliferation rate of MC3T3-E1 cells on 1d,3d,5d,and 7d was gradually increased,but there was no statistic significant(P>0.05).The biomechanical examination of the interface bone tissue specimens of the two groups of rabbits after 12 weeks of surgery showed that the failure load and stiffness of the observation group were significantly higher than those of the control group,and the difference was statistically significant(P<0.05).Biomechanical Levels of Healing Interface Bone Tissue Specimens in 12 weeks after surgery for two groups of white rabbits show that the failure load,stiffness of observation group were higher than that of control group,the difference was statistic was significant(P<0.05).The fracture healing scores of observation group on 4th,8th,and 12 th weeks were higher than that of control group,the difference was statistic significant(P<0.05).Conclusion: The silk fibroin/graphene oxide composite membrane has good compatibility and biomechanical properties,and it can promote the healing of long bone nonunion.
