| Objective: to investigate the epidemic status of Hemorrhagic Fever with Renal Syndrome(HFRS)in Hebei province in 2017,to investigate the rat density and HV carrying rate of the National surveillance site in Hebei province,to explore the interaction and molecular mechanisms of the Seoul Virus(SEOV)and cell autophagy,and provide the gist for the study of SEOV pathogenesis and the prevention and control of HFRS.Methods:1 Using the description epidemiology method and statistical analysis to analyze the information of HFRS cases in Hebei province in 2017 from the Chinese disease prevention and control information system.2 Using the night trapping method to investigate the rat density and the rat species were identified at the same time.Direct immunofluorescence was used to detect the HV antigen of rat lung,and calculate the HV carrying rate.3 The changes of the autophagy level induced by SEOV infection were observed by means of plasmid transfection,immunofluorescence,Western blot and real-time fluorescence quantitative RT-PCR.4 Rapamycin,3-MA,E64 D were used to induce or inhibit autophagy and observe the changes in SEOV’s replication and release levels,using untreated cells infected by SEOV as Control.Results:1 In 2017,456 cases of HFRS were reported in Hebei province.Cases were occured every month.Spring(march-may)was the peak period of the onset of the disease,which accounts for 40.9% of the year.Qinhuangdao and Tangshan reported the highest number of cases,accounting for 44.3% and32.2% respectively.50-55 age group had the most Cases,and the incidence of male was significantly higher than that of female.The occupation distribution of cases was mainly farmers(71.9%).2.In 2017,a total of 792 rats were captured,and the average density of rodents was 1.75% while the average HV carrying rate of rats was 3.51%.The HV carrying rate of rats in Qinhuangdao city was the highest with 11.83%.3.Compared with the control,SEOV infection of Vero E6 cells can induce the exogenous RFP-LC3 protein in cells and it increased over time.Intracellular LC3-II protein expression was up-regulated.4.Compared with the control,promoting autophagy can significantly improve the level of intracellular virus replication.Similarly,inhibition of autophagy can significantly reduce the level of intracellular virus replication.Conclusion:1.In comparison with historical monitoring data,the incidence of HFRS in Hebei province in 2017 is at a lower level,and the epidemiology characteristics has not changed significantly.The HFRS control and prevention in northeast areas of Hebei province prevention still need to be paid close attention to.2.SEOV infection can induce autophagy in Vero E6 cells,m RFP-LC3 and endogenous LC3-IIexpression were significantly enhanced.3.Promoting autophagy can enhance SEOV replication.Autophagy inhibiting can decreass thereplication of SEOV. |
