| Background:A large number of studies have shown that Chinese herbal medicines have antioxidant effects.Semiaquilegia adoxoides is a commonly used traditional Chinese medicine,has the effects of clearing away heat,detoxifying,swelling and diuresis,and diuresis and dripping.Currently,the research is only the analysis of its chemical composition,but the specific function of its specific chemical composition has not yet existed.In-depth studies and phase studies have shown that the n-butanol extract of Semiaquilegia adoxoides has protective effects on human lens epithelial cells against oxidative stress.Objective:In this study,we used the response surface method to optimize the ultrasonic extraction of total alkaloids from Semiaquilegia adoxoides(DC.)Makino,and obtained the conditions for the optimal extraction of total alkaloids from Semiaquilegia adoxoides and the in vitro antioxidation of total alkaloids..In addition,the anti-aging effects of Semiaquilegia adoxoides on D-galactose-induced subacute aging model mice were studied,and the mechanism of anti-aging of Semiaquilegia adoxoides was discussed.Methods:Using Design Expert 8.0.6,the response surface test of ethanol concentration,liquid-to solid ratio,ultrasonic time and ultrasonic power and to investigate the effects of DPPH method,ABTS method and total reduction ability on in vitro antioxidant.Kunming mice were randomly divided into 6 groups.Except the normal group,500 mg·kg-1D-galactose was administered subcutaneously to the neck and back of the rats,once a day for 4 weeks to induce senescence-like changes.At the same time The rats were given 600,300 and 150 mg·kg-1 or 100 mg·kg-1of vitamin E(positive control group)and normal saline(normal group)by intragastric administration once daily,For 4 consecutive weeks,mice were weighed once a week.After 3 weeks of continuous gavage,the mice behavioral indexes were examined,and the water maze test was used to measure the memory ability of mice.After 4 weeks,the activities of antioxidantenzymessuchassuperoxidedismutase(SOD),malondialdehyde(MDA),catalase(CAT)and glutathione peroxidase(GSH-Px)index of aging.Results:The optimum extraction conditions were as follows:ethanol concentration was 86%,liquid-to solid ratio was 37:1 mL·g-1,Ultrasound time of 32 min,ultrasonic power of 140W.The IC500 of DPPH free radicals was 0.631 mg·L-1,the IC500 value of scavenging ABTS free radical was0.747 mg·L-1.The results showed that the number of mice in the model group and the number of hindlimb standing significantly reduced,the incubation period is prolonged,the number of traverses decreased.The contents of MDA in senescent mice were significantly increased,and the activities of SOD,CAT and GSH-Px in serum,liver and kidney were decreased to some extent,compared with normal mice(P<0.05)The decoction of 600 mg·kg-1 g and 300 mg·kg-1 all increased the activities of SOD,CAT,GSH-Px in the serum,liver and kidneys,and decreased the content of MDA in the senescent mice compared with the model group Difference(P<0.01).Anddecoction 150 mg·kg-1 can increase serum SOD,CAT,liver SOD,kindey CAT activity,reduce serum and liver MDA content.Conclusion:Indicating that the optimum process conditions were stable and feasible,and the concentration of active and total alkaloid was apparent dose-effect relationship.SA can improve the learning and memory ability of D-galactose-induced aging mice,and delay the body senescence of D-galactose-induced model mice. |
