PURPOSE: For the early detection and diagnosis of cervical lymph node metastasis,we developed a new multifunctional nanocarbon SW4.This study aims to investigate the application value of cervical lymphatic imaging enhanced by submucosal injection of SW4.It provides a new way of thinking for judging the nature of cervical lymph nodes and the multifunction of contrast agents.METHODS:0.2ml C/Gd nanocomposite was injected into the submucosa of the bilateral tongue in 12 healthy Zealand white rabbits and then massaged the injection site for 30 seconds.MR images were obtained before injection and 5,10,15,20 and 40mins after injection by using 3D enhancement magnetic resonance lymphography.The signal intensities were measured to calculate the enhancing rates(E %)and the signal enhancing rates—time curve was drawn before and after enhancing of cervical lymph nodes.The data were statistically analyzed by using SPSS17.0 software package.Blood chemistry and blood routine were measured,and histopathologic examination of liver and kidneys was done before and 4w after the experiment.RESULTS: After injection,C/Gd nanocomposite was rapidly absorbed into the cervical drainage area.The lymph nodes and lymphatic vessels were strengthened significantly;however,the blood vessels were not displayed at the same time.The enhancing rates of the cervical lymph nodes reached a peak at 10 mins after injection,and the best enhancing effect was between 5mins and 20 mins,and would not be able to distinguish with that of the surrounding tissues after 40 min.There were no obvious different of Blood Chemistry and blood routine before and 4w after the injection.The structure of liver,kidney did not change significantly with pathological examination after the experiment.All of the rabbits in each group have no adverse reactions after interstitial MR lymphography.CONCLUSIONS: As IMRLG contrast agent,C/Gd nanocomposite can characteristically display the cervical lymph node morphology and lymphatic shape effectively with a small doses.There was no obvious side effect,.but a significant lymphoid specificity.It can provide new way or evidence for the diagnosis of cervical lymph node metastasis.At the same time,its modifiable surface and photothermal conversion capability lay a foundation for the study of tumor targeted drug delivery and photothermal therapy.
