Objective:To observe the effect traditional Chinese medicine(TCM)of BuShenHuoXue on the injury optic nerve rat model of chronic elevated intraocular pressure(EIOP)and preliminarily explore its mechanism.Methods:By unilaterally cauterizing 3 episcleral vessels, establish the rat model of chronic EIOP.30 rats were divided into 3 groups randomly:model group, treatment group, and normal control group. After given TCM drugs of BuShenHuoXue or normal saline for 8 weeks, the rats were put to death.To observe the effection of BuShenHuoXue method on the EIOP rat model's intraocular pressure(IOP), the content of myelin sheath of optic nerve fibre, Pathological changes of optic nerve and expression of brain-derived neurotrophin factor(BDNF).Results:Unilaterally cauterizing episcleral vessels increased IOP of the rat modle obviously(P<0.01).At the end of the experiment, the level of IOP was 2.5-3 times than Before modeling; the effects of BuShenHuoXue on the model optic nerve injured rat of chronic EIOP as follows:slightly lower IOP; repaired injury optic nerve; increased the content of myelin sheath of optic nerve fibre:improved the expression of BDNF of optic nerve on the EIOP rat model.Conclusion:This model is simple and stable, and moderately elevated IOP, which can last at least 2 months. TCM of BuShenHuoXue can protect the visual function on the rat model of chronic EIOP.The effects mainly effects as:Slightly lower IOP; repaired injury optic nerve;increased the content of myelin sheath of optic nerve fibre; improved the expression of BDNF of optic nerve on the EIOP rat model.
