Objective:To observe the regulatory mechanism of GDNF in cerebral ischemia reperfusion rats,through the expression level of IRAK,TAK1 and GDNF at different time points in rats after cerebral ischemia reperfusion injury was detected by the detection of the "Shuigou","Neiguan","Weizhong","Sanyinjiao".And to discuss the molecular mechanism of regulating GDNF expression in detail.Methods:A totle of 120 Male adult SD rats were randomly divided into five groups,the electric acupuncture group and inhibitors 5 groups,each group according to 3,7,14,28 days after treatment was divided into four subgroups,each subgroup have 6 rats.Model group,electroacupuncture group and inhibitor group were used to prepare the focal cerebral ischemia-reperfusion model in the right cerebral artery by referring to Longa modified line suppository method.After modeling,the rats were judged by neurological function to determine whether the model was successful.3days after the building successfully,the curative group selection "off" in "ditch" "bilateral" "three vaginal intercourse",stimulation intensity to the needle handle slightly shaking,no violent struggle for degrees,of rats every 30 min,1 time/day;The inhibitor group was injected with NF-?B inhibitor PDTC,and the electroacupuncture group was operated with the electroacupuncture group.In the same time period,the remaining group of rats were grasping and fixed,and no operation was performed.In the treatment of 3,7,14,28 days after the point in time,after observed between groups of neural behavior of rats,randomly selected from 6 to be put to death in the brain,extraction of brain ischemia in rats and with brain tissue,and application of real-time fluorescent quantitative PCR technique to detect IRAK,TAK1,GDNF mRNA expression level.Results:1 Completion of cerebral ischemia model: the total number of rats in this experiment was 120,and 85 mice were selected,and the success rate was 70.8%.Are eliminated in rats,building 12 only losers,victim 23(3 died of anesthesia,only fivedied of intraoperative improper operation,15 died of postoperative cerebral ischemia injury is overweight,infection or bleeding in the brain).2 Neurological function score of rats: In the blank group and the sham operation group,there was no neurological impairment,the score was 0,and the sham operation group was no more significant than the blank group,with no statistical significance(P <0.05).Before treatment,the score difference was statistically significant(P<0.05)compared with the blank group in the model group,the electroacupuncture group,the inhibitor group and the blank group.Each point in time after treatment,the curative group,the inhibitor group compared with model group,neurobehavioral scores tend to decrease gradually,3,7,28 days,the difference statistically significant(P<0.05).3 The comparison expression of IRAKmRNA of cerebral ischemic semidark band tissue:Compared with the blank group,the expression level of IRAKmRNA in the model group was significantly increased in the third day after treatment(P < 0.05).Compared with the model group,the expression level of IRAKmRNA was significantly reduced in the electroacupuncture group during the treatment of the 3rd day,the 14 th day was significantly increased(P<0.05),and the inhibitor group was significantly reduced on the third day(P < 0.05).Compared with the electroacupuncture group,the expression level of IRAKmRNA in the treatment of 3,14 and 28 days was significantly reduced(P<0.05).4 The comparison expression of TAK1 mRNA of cerebral ischemic semidark band tissue:Compared with the blank group,the expression level of TAK1 mRNA in the model group was significantly increased in the third and 14 th days(P < 0.05).Compared with the model group,the expression level of TAK1 mRNA in the electroacupuncture group was significantly reduced(P<0.05)in the treatment of 3,7and 14 days.Compared with the electroacupuncture group,the expression level of TAK1 mRNA in the treatment of 3 and 28 days was significantly reduced(P<0.05).5 The comparison expression of GDNFmRNA of cerebral ischemic semidarkband tissue:Compared with the blank group,the expression level of GDNFmRNA in the model group was significantly increased in the third,7th and 14 th days(P<0.05).Compared with the model group,the expression level of GDNFmRNA in the electroacupuncture group was significantly increased(P < 0.05),and the inhibitor group was significantly down-regulated in the third,7th and 14 th days.Compared with the electroacupuncture group,the expression level of GDNFmRNA was significantly reduced at each time point(P<0.05).Conclusion:Electric acupuncture stimulate the“Shuigou”,“Neiguan”,“Weizhong”,“Sanyin jiao”acupoints to reduce the expression level of TLR2?NF-?B?TRAF6 on the TLR2/NF-?B signaling pathway,and improve the functional rehabilitation of injured nerve.The mechanism of action may be related to the activity of inhibiting the TLR2/NF-?B signaling pathway after the reduction of IRAK and TAK1.
