Globba racemosa Smith is the Zingiberaceae family Globba L plants,which is a herb for many years,It is mainly distributed in southern and southwestern provinces of China.Born in the damp place under the forest,4001300 m above sea level,its gas is slightly,tasteless,and slightly sweet.The roots and fruits of G.racemosa Smith medicinal herbs have functions such as stomach,anti-inflammation,guilty,lungs and cough,and Shujin Huoluo.They are mainly used in the treatment of gastritis,dyspepsia,acute and chronic nephritis,acute edema,uterine bleeding,and labor.Injuries,coughs,wheezing,bloating,rheumatism,and other symptoms have a significant effect.However,due to the lack of relevant statutory quality standards,G.racemosa Smith Herbal Medicine is extremely limited in its in-depth development and utilization.In order to change this situation,this project took G.racemosa Smith medicinal materials as the research object,and guided the technical requirements of the 2015Chinese Pharmacopoeia as the guide to carry out relevant research work such as quality control standards for G.racemosa Smith medicinal materials.The research work and results are as follows:1.Study on Volatile Chemical Components of G.racemosa SmithThe essential oil from the roots,stems and leaves of G.racemosa Smith was obtained by steam distillation and analyed by the gas chromatography mass spectrometry?GC-MS?technology,the relative content was determined by area normalization.The results show:fifty-nine components were identified from the roots,stems and leaves of G.racemosa Smith.Thirty-two components were identified from the roots,in which accounting for 91.74%of total volatile oil.The main components of the volatile oil in roots are?-pinene?14.20%?,Linalool?14.06%?,?-Terpinene?12.64%?,camphere?11.67%?.Twenty-six components were identified from the stems,in which accounting for 56.33%of total volatile oil.The main constituents of the volatile oil of stems are Ambrial?12.00%?,d-Nerolidol?7.67%?.Twenty-seven components were identified from the leaves,in which accounting for 74.15%of total volatile oil.The major components of volatile oil from leaves are?-?-?-Elemene?25.83%?,d-Nerolidol?10.89%?.2.Study on extraction process of total saponins from G.racemosa SmithThe saponins are one of the main components of G.racemosa Smith,this section of the experiment conducted an extraction pretreatment study and evaluated the antioxidant activity of the extracts.On the basis of single factor experiment,adopting3-factor 3 level response surface test was used to investigate and study.The optimal pretreatment parameters for the total saponins of roots of G.racemosa Smith were:the ratio of liquid to material was 1:27.6(g·mL-1),the particle sizes of sample were 37.48mesh and the ultrasonic extraction time was 51 min.According to this process,the total saponins in the roots of G.racemosa Smith were extracted in different habitats,the contents of total saponins in Wuchuan County and Wuchuan County reached 16.88mg·g-1 and 16.92 mg·g-1,respectively.Antioxidant analysis was performed using DPPH·and ABTS+·.The results showed that the concentration of extracts of root of G.racemosa Smith was positively proportional to the antioxidant activity,the radicals of the roots of G.racemosa Smith is from different regions had different radical scavenging rates,and the scavenging rate and total saponin content were strong and weak.The dose-effect relationship showed that its antioxidant capacity was related to the total saponin content.3.Study on fingerprint of G.racemosa Smith medicineThe HPLC fingerprint-diode array detector was used to establish the fingerprint of G.racemosa Smith,which was analyzed using the“Chinese herbal medicine chromatographic fingerprint identification degree evaluation software”?2012 edition?.A total of 15 common peaks of G.racemosa Smith were identified,the acquaintance degree of the fingerprints of the G.racemosa Smith samples was all above 0.9,indicating that the quality of the G.racemosa Smith medicinal materials is stable and controllable,and it can be used as a quality control studies.According to the results of systematic cluster analysis,G.racemosa Smith medicinal herbs from 13 batches different origins can be divided into 2 categories:A and B,it is classified as Category A in the Qiandongnan and Qiannan areas,and classified as Category B in the Tongren and Zunyi regions.4.Study on the activity of G.racemosa Smith extracts in vitroIn this part of the experiment,the extracts from the roots of G.racemosa Smith made with petroleum ether,75%ethanol and water were used to conduct in vitro antioxidant activity,acetylcholinesterase inhibitory activity,and alpha-amylase inhibitory activity.The results showed that the extracts prepared made with three solvents showed some differences in the three in vitro experiments.For the same origin,the antioxidant capacity of the sample extracts are as follows:petroleum ether extracts>75%ethanol extracts>aqueous extracts;the order of inhibitory activity of acetylcholinesterase in the sample extracts are:75%ethanol extracts>The petroleum ether extracts>water extract;the order of inhibitory activity of the?-amylase of the sample extracts are:water extracts>75%ethanol extracts>petroleum ether extracts.All three solvent extracts have different biological activities,which provide some reference and scientific basis for the separation and purification for roots of G.racemosa Smith.5.Study on quality standard of G.racemosa SmithAccording to the relevant technical regulations and guidelines of the 2015 edition of the Chinese Pharmacopoeia,the water content,total ash content,acid-insoluble ash content,heavy metal content,and organic pesticide residue content in 13 different origins of G.racemosa Smith medicinal herbs were systematically determined.With?-sitosterol as control,establishment of 13 batches of different places G.racemosa Smith medicinal thin layer identification,the total saponin content was determined by UV-Vis spectrophotometry and study on antioxidant activity.With reference to ancient books and modern clinical research,the description of the sexual taste of G.racemosa Smith,description of function,dosage,precautions and storage are described in writing.Based on the above research contents,according to the technical guidelines of the2015 Chinese Pharmacopoeia on the quality standards of medicinal materials,draft the draft quality standard of G.racemosa Smith medicinal materials and attach drafting instructions.It is expected that through this scientific research,the research results formed will be included in the latest edition of the“Quality Standards for Chinese Medicinal Materials and Ethnic Medicinal Products in Guizhou Province”,which will make the G.racemosa Smith Herbal Medicine“accommodated”in terms of production,sales,and creation of new drugs.
