Isodon ternifolia?D.Don?Kudo,a number of Isodon,belonging to rabdosia family,is widely distributed in Guangxi,Guangdong and Yunnan Provinces of China.Isodon ternifolia?D.Don?Kudo has been used in traditional medicine,I.ternifolia is often used for the treatment of conditions including traumactic injury,flu,toothache and snake bite.Previous phytochemical studies on I.ternifolia has led to the isolation and identification of diterpenoids,flavonoids,and several other constituents.Isodon exhibited diverse biological activities such as anti-inflamma-tory and cytotoxic.Diterpenoids are the main active components of Isodon.In this study,the chemical composition and anti-inflammatory activity of EtOAc and n-Bu OH part from the roots of I.ternifolia were studied by chromatographic methods and spectroscopic methods.In the dissertation,the EtOAc and n-BuOH part from the roots of I.ternifolia was isolated using silica gel column chromatogaphy?CC?,MCI CC,Sephadex LH-20 CC,recrystallization,semi-preparative CC and preparative hign performance liquid chromatograph?HPLC?CC.25compounds were isolated and identified by spectrometric data of 1H NMR,13C NMR and MS.These compounds were identified as folloes:3-carboxy formate-6,7-dihydroxy-1-?3?,4?-dihydro-xyphenyl?-naphthalene?1?,3-carboxy-6,7-dihydroxy-1-?3?,4?-dihydroxyphenyl?-naphthalene?2?,1-acetatexyl-2e,6e-dipiperonyl-3,7-dioxabcyclo[3,3,0]-octane?3?,2-?3,4,5-trimethoxyphenyl?-6-?4-hydroxy-3-methoxyphenyl?-3,7-dioxabicyclo[3.3.0]octane?4?,syringyl-glycerol-?-syring-aresinol ether 4",4-m-di-O-?-D-glucopyranoside?5?,clemastanin A?6?,3-?3,4-dihydroxyphenyl?acrylic acid 1-?3,4-dihydroxyphenyl?-2-methoxycarbonylethyl ester?7?,oxyneolignans A?8?,19-hydroxytotarol?9?,?-?-lambertic acid?10?,6?,11?,15?-triacetoxy-7?-hydroxy-7?,20-epoxyentkaur-16-ene?11?,longikauf A?12?,maoyecrystal F?13?,2?-hydroxy oleanolic acid?14?,tormentic acid?15?,myricetin 3-neohesperidoside?16?,3,4-dimethoxy-1-hydroxylphenyl-O-?-D-glucopyranoside?17?,2,6-dimethoxy-p-hydroquinonel-O-?-D-glucopyranoside?18?,3,4,?-trihydroxyl-methyl phenylporpionate?19?,erythro-l-c-syringyl-glycerol?20?,E-thyltrans-p-coumaric hydroxyl cinnamate?21?,caffeic acid?22?,3,4-dihydroxyphenacyl alco-hol?23?,protocatechuic acid?24?and syringic acid?25?.According to the data-base of SciFinder Scholar,compound 1 was new compound,compounds 2,4,5,8,16,17 and 18 were firstly isolated from Isodon.The inhibition of NO production by LPS-induceds in RAW264.7 cells of six compounds was evaluated.The results showed that compounds 1,2,3 and 5 could inhibit the NO production in RAW264.7 cells.The most active compound 1 exhibited best inhibition of NO production with IC50 values was 9.98mM.The chemical constituents and anti-inflammatory activities of rabdosia trifoliata were further revealed,which provided a theoretical basis for the further development of Isodon ternifolia.
