Experimental Study Of The Apoptosis And Its Mechanism Of ?-Elemene On Children's Neuroblastoma Cells | Posted on:2019-06-19 | Degree:Master | Type:Thesis | Country:China | Candidate:Y Yang | Full Text:PDF | GTID:2394330566982484 | Subject:Academy of Pediatrics | Abstract/Summary: | PDF Full Text Request | Objective: The inhibitory effect of ?-elemene on the growth of Neuroblastoma(Neuroblastoma,NB)and its possible mechanism were investigated with neuroblastoma cell lines SH-SY5 Y and SK-N-DZ,which provided a theoretical basis for the clinical application of the neuroblastoma.Methods:(1)NB cells SH-SY5 Y and SK-N-DZ were cultured,and logarithmic growth phase cells were treated with different concentrations of?-elemene for different times in vitro,then cell viability was measured by using a cell counting kit-8 assay.(2)After treated with ?-elemene for 24 h,cell apoptosis,cell cycle and the incorporation of BrdU was measured by flow cytometry.Hoechst 33342 staining was used to detect the apoptotic bodies.And the expression of caspase-3,caspase-8 and caspase-9 was measured by Western blot.Results:(1)The inhibitory effect of ?-elemene on NB cells was showed in a dose-and time-dependent manner.(2)After treated with?-elemene for 24 h,the rate of early apoptosis of NB cells was increasedcompared with the control group(P < 0.05).(3)Apoptotic bodies were increased obviously compared with the control group(P < 0.05).(4)There was no significant difference in the proportion of G0/G1,S and G2/M in each cell cycle(P > 0.05).(5)The incorporation rates of BrdU had no significant difference(P > 0.05).(6)The expressions of caspase-3 and caspase-9 protein were increased significantly(P < 0.05),but there was no significant difference in the expression of caspase-8(P> 0.05).Conclusion: ?-elemene can inhibit the viability of NB cells,which is due to induce apoptosis by up-regulating the expression of caspase-3 and caspase-9,and doesn't influence cell cycle and proliferation. | Keywords/Search Tags: | ?-elemene, neuroblastoma, cell apoptosis, caspase-3, caspase-9 | PDF Full Text Request | Related items |
