Oxidative stress refers to a series of damages caused by the imbalance of the redox system in the body,which is manifested as excess free radicals in the body,active intermediates of reactive oxygen species cannot be promptly removed,resulting in the oxidative damage of the body.Oxidative damage can lead to a series of diseases of the nervous system,such as Alzheimer's disease(AD),Parkinson's disease(PD)and other diseases.Antioxidant treatment to some extent points out new directions for the treatment of these diseases.Cynanchum saccatum is a plant belonging to the Apocynaceae,which is mainly distributed in Tibet,Yunnan,and Sichuan.Early studies found that it had anticonvulsant effects and was similar to the pharmacological effects of Cynanchum otophyllum Schneid,but its antioxidative effects were less studied.Therefore,as a new natural antioxidant,it is attracting attention from all walks of life.Curcuma aromatica Salisb.is a turmeric plant belonging to the Zingiberaceae.It is mainly distributed in the southeast and southwest regions of China.Its rhizome is included in the Chinese Pharmacopoeia as a traditional Chinese medicine,it has the effects of promoting qi stagnation,cooling blood and breaking blood stasis.Previous pharmacological studies have shown that the same plant of Zingiberaceae has good antioxidant and neuroprotective effects,so the antioxidant effect of Curcuma aromatica Salisb.as a natural active substance is well worth looking forward to.In this paper,the oxidative damage model of PC 12 cells induced by H2O2 was used to evaluate the antioxidant activity of 29 compounds isolated from Cynanchum saccatum and 36 compounds isolated from Curcuma aromatica Salisb..and to explore the mechanism of its protective effect on PC12 cells.Studies have shown that the monomer compounds extracted from the two plants have good anti-oxidation effect and can effectively reduce the damage of PC 12 cells induced by H2O2.They can activate the Nrf2-ARE pathway and regulate the expression of HO-1 and NQO1.By regulating the activities of antioxidant enzymes such as GSH,SOD,CAT,and LDH,reducing the intracellular levels of ROS and Ca2+ and decreasing the apoptosis rate of cells to achieve the antioxidation effect.The research work of this paper provides a certain preliminary basis for the discovery of natural antioxidants,and also provides a reference for the rational development of the above two kinds of medicinal plants.
