Aim:This study was to observe the effect of Danzhi Compound Decoction on general conditions ? thyroid hormone functions ? Pathological sections and the expression of thyroid Caspase-3?Bcl-2?Bax of Graves' Disease Mice.To provide theoretical support for the traditional Chinese medicine curing Graves' Disease,and point out that the therapeutic mechanism may be related to the expression of Caspase-3?Bcl-2?Bax in thyroid.Method : Randomly divided healthy female Balb/c mice into blank group and model group.Using Adenovirus to make the model of Grave Disease mice.After modeling success,randomly divided the model group into 5 groups: model group(MG)?western medicine group(WMG)?Danzhi Compound Decoction low dose group(LDG)?Danzhi Compound Decoction middle dose group(MDG)?Danzhi Compound Decoction high dose group(HDG),And give them the corresponding drug gavage.The course of treatment was six weeks.Observing and monitoring the general situation?diet and drinking?body temperature and body weight of mice during the treatment.Six weeks after,took blood by remove eyeballs to exam thyroid hormone functions in Elisa method,removed the thyroid to exam the expression of thyroid Caspase-3?bcl-2?Bax in Western Blot,and thyroid histology were observed by using HE staining.Result : 1.After 6 weeks' immunization,compared with the blank group,the model mouse show high spirit?restless?aggressive.Diet and water increased,anus temperature increased,body weight slowly growed;The level of TSH in Model Group was significantly lower than the Blank Group while the levels of T4,T3 and Trab were significantly higher;Thyroid HE staining was diffuse toxic goiter.2.After 6 weeks' treatment,compared with the model group,the diet of MDG was extremely significantly increased(P <0.01);the diet of the WMG and the LDG were significantly increased(P <0.05);the water drink of WMG ? MDG ? HDG were extremely significantly increased(P <0.01);anus temperature of WMG and TCMgroups has a decrease trend(P> 0.05);MDG and HDG significantly increased body weight(P<0.01).3.After 6 weeks' treatment,the serum levels of T3 in the WMG and MDG were significantly decreased(respectively,P <0.01,P<0.05);The serum levels of T4 in the WMG and TCM groups were extremely significantly decreased(P <0.01);The serum levels of Trab in the WMG and MDG were significantly decreased(P <0.05);The serum levels of TSH in the WMG and MDG were extremely significantly increased(P<0.01),The serum levels of TSH in the HDG was significantly increased(P <0.05).4.After 6 weeks' treatment,the expression of thyroid Caspase-3 in each groups were extremely significantly decreased(P <0.01);The expression of thyroid Bcl-2 in WMG ? MDG and HDG were extremely significantly increased(P <0.01);The expression of thyroid Bax in each groups were extremely significantly decreased;Bcl-2/Bax in WMG and HDG were significantly increased(respectively,P<0.01,P<0.05)?5.The expression of Caspase-3 and Bax was positively correlated with the levels of T3,T4 and Trab,and negatively correlated with the level of TSH.The expression of Bcl-2 was negatively correlated with the levels of T3,T4 and Trab,and positively correlated with the level of TSH.The ratio between Bcl-2 and Bax was negatively correlated with T3,T4 and Trab levels,and positively correlated with TSH level.Conclusion:1.The model of Graves' disease mice was successfully established in this experiment,Danzhi Compound Decoction and Methimazole both have positive effection on the serological indicators of Graves' disease mouse,but the efficacy of methimazole is better than Danzhi Compound Decoction.2.Graves' disease mouse exist abnormal apoptosis,the degree of abnormal apoptosis and Graves' disease was positively correlated.3.Danzhi Compound Decoction and methimazole are able to down regulate the expression of Caspase-3 and up regulate the expression of Bcl-2.In those dose groups,the effect of the high dose group was the best.4.Caspase-3?Bcl-2?Bax may be involved in the regulation of thyroid apoptosis.
