Objective: To observe the hyperthyroidism and thyroid function in mice liver oxidative stress,to understand the effect of Danzhi compound on mouse thyroid function and liver function,and to observe the effects of Danzhi compound on mice liver oxidative stress,clear therapeutic effect of Danzhi compound on mice with hyperthyroidism.Methods: The animal model of GD Ad-TSHRa-289 immune BALB/c mice,mice were randomly divided into normal control group,model group,Danzhixiaoyao powder compound is low,middle and high dose intervention group,the intervention group of methimazole during administration;observe the general condition and body weight,diet and water change,after 6 weeks,the serum and thyroid tissue and liver tissue.With the competition method to detect serum TSH,T3,T4 content,radioreceptor assay to detect the concentration of serum TRAb,thyroid tissue HE staining to verify the curative effect of Danzhi Xiaoyao Powder compound.olorimetric method was used to detect serum AST,ALT,ALP in liver tissue of mice were determined in antioxidant enzymes SOD,CAT,DGSH-PX activity,to investigate the effects of Danzhi compound on mice with hyperthyroidism thyroid function,liver injury and liver oxidative injury.Results:1.Adenovirus immunization for 6 weeks,the model group TSH was significantly lower than the blank group,T4,T3,TRAb was significantly higher than the blank group.TSH,T4,T3 and TRAb in the western medicine group were better than those in the treatment group.2.6 weeks after the adenovirus immunization,mice model AST,ALT,ALP was significantly higher than the control group,the comparison between the treatment group and compound Danzhi AST of high dose group was significantly lower than that in the western medicine group,low dose group,compound Danzhi AST lower than western medicine group;compound Danzhi ALT of high dose group was significantly lower than that in the western medicine group,low compound Danzhi middle dose group ALT was lower than that of Western medicine group;Danzhi compound in the high dose group of ALP was significantly lower than that of the western medicine group,low dose group of compound Danzhi ALP was lower than that of western medicine group.3.adenovirus immunization for 6weeks,the model group MDA,SOD,GSH-PX was significantly higher than the blankgroup.The comparison between the treatment group and compound Danzhi MDA in middle and high dose groups was higher than control group,low dose group of compound Danzhi MDA was significantly higher than the control group;Danzhi compound in the high dose group of SOD was significantly lower than that of the western medicine group,low dose group of compound Danzhi SOD lower than western medicine group;western medicine group GSH-PX was significantly lower in low dose group of compound Zhi Yu Dan.The western medicine group GSH-PX was higher than that of the high dose group,western medicine group GSH-PX lower dose group: compound.4.MDA,SOD,GSH-PX and TSH negatively correlated with the level of MDA,SOD,GSH-PX;and T4 levels were positively correlated;SOD,GSH-PX and T3 levels were positively correlated;ALT and AST were negatively correlated with the level of TSH,AST and T4;ALT was positively.5.HE staining,the model mice of thyroid pathological manifestations of thyroid follicular epithelial cells increased,vacuolar degeneration,tissue vascular invasion,follicular morphology was irregular,glial less,there is a small amount of inflammatory cells;western medicine group thyroid pathology showed edema,thyroid epithelial cell shedding,part of colloid in follicles concentration,vascular tissue invasion,infiltration of inflammatory cells;and low doses of mice thyroid pathology showed thyroid epithelial hyperplasia in different degrees in compound Danzhi irregular shape,follicular tissue,inflammatory cell infiltration.There was no significant abnormality in thyroid pathology in mice.6.In the model group,the pathological features of the liver showed that the surface of the liver cells edema,part of the hepatic sinus expansion,there were inflammatory cell infiltration in the District,the nucleus was round,oval,the size of the liver cell necrosis.In the western medicine group,the pathological features of the liver showed that the hepatic tissue structure was clear,the nuclei were round and oval,some of the nuclei were enlarged,and some of the hepatic sinusoids were dilated.The liver pathology showed compound Danzhi liver tissue structure is clear,the nuclei are round,occasional swelling of liver cells.Conclusion: 1.Graves mouse model was successfully established,and the therapeutic effect of methimazole on compound Danzhi hyperthyroid mice were,but curative effect is better than that of compound Danzhi methimazole.2.Hyperthyroid mice had liver injury,and the degree of liver injury was positively correlated with hyperthyroidism.3.Compound Danzhi have therapeutic effect and methimazole forliver injury in hyperthyroidism,but the curative effect is better than that of compound Danzhi methimazole.4.There was a positive correlation between the degree of oxidative damage and the degree of hyperthyroidism.5.Liver oxidative damage has the treatment result of 5methimazole and Danzhi compound caused by hyperthyroidism,Danzhi compound is better than the high dose of methimazole,Danzhi compound dose to improve oxidative damage is the best treatment.
