Objective:To analyze the effect of electroacupuncture,moxibustion and gusha on the effect of“Huantiao”(GB30)and“YangLingQuan”(GB34)in the rats of CCI model,and to explore the biological mechanism of electroacupuncture,moxibustion and scraping treatment for sciatica was studied.Methods:12 rats of clean grade SD(Sprague Dawley)were divided into electroacupuncture group and moxibustion group with reference random number table,6rats in each group,self control,intervention 1 times and perfusion,and the effect of electroacupuncture and moxibustion on the effect of normal SD rodent area.30 clean male SD rats were divided into blank group,false model group,model group and 10 rats in each group.The mechanical stimulation and thermal radiation stimulation of the left foot of rats were carried out at first,2,3,4 and 5 days,and their behavioral changes were observed.After the success of the model,35 clean male SD rats were randomly divided into blank group,model group,electroacupuncture+model group,moxibustion+model group,scraping+model group,7 rats in each group.After the model,third days after the model and1 times of intervention,the left foot of the rat was stimulated by mechanical stimulation and heat radiation stimulation.The effect of moxibustion and scraping on the area effect of sciatica in rats.The intervention site selected the left hind limb of the rat,“Huantiao”(GB30)and“YangLingQuan”(GB34)point,and the intervention methods were divided into groups.The model of CCI rats was used in the model.The local skin tissue of the acupoint area was detected by 4%formaldehyde perfusion,and the statistical principle was used to analyze the related data.Results:1.Compared with the control group,the expression of five hydroxytryptamine(5-HT)and histamine(HA)in the local skin tissues of healthy SD rats after moxibustion increased significantly.The number of 5-HT and HA positive cell markers in 10 times microscope was significantly higher in moxibustion group than in control group(*P<0.05);Compared with the control group,the expression of substance P(SP),calcitonin gene related peptide(CGRP),five hydroxytryptamine(5-HT)and histamine(HA)in the local skin tissue of“YangLingQuan”(GB34)point after electroacupuncture was significantly enhanced in SD rats.the length of SP and CGRP positive fibers was counted under 20 times of microscope.The markers of 5-HT and HA positive cells were counted under 10 times microscope.The results showed that the electroacupuncture group was significantly higher than that of the control group.The difference was statistically significant(*P<0.05);2.After the CCI model was prepared,the rats appeared limp,with mild hallux valgus.There was no significant difference between the mechanical pain threshold and the thermal pain threshold of the blank group,the fake model group and the model group before making the model.The mechanical pain threshold and the thermal pain threshold in the blank group and the false model group had no significant changes,and the model group decreased significantly(▲*P<0.05),and decreased to the lowest and continued to fourth,fifth days at the third day.3.Behavioral changes before and after electroacupuncture,moxibustion and guasha therapy:Before modeling,blank group,model group,electro acupuncture+model group,moxibustion+model group,scraping+model group,there was no significant difference between groups;Compared with the model before the model,the mechanical pain threshold and the thermal pain threshold of the third sky white group had no obvious changes.The model group,the electroacupuncture+model group,the moxibustion+model group,the scraping+model group decreased significantly the mechanical pain threshold and the thermal pain threshold,which had statistical significance(△#P<0.05);Compared with the third days after the model,the mechanical pain threshold and the thermal pain threshold of the blank group and the model group were not significantly changed after the intervention.Themechanicalpainthresholdandthethermalpainthresholdinthe electroacupuncture+model group,the moxibustion+model group,the scraping+model group increased significantly,and the mechanical threshold difference was statistically significant(*P<0.05).4.The epidermal layer and dermis of the“Huantiao”(GB30)and“YangLingQuan”(GB34)area have 5-HT positive markers,which form a discontinuous cell band parallel to the epidermis in the epidermis and the superficial epidermis.In the dermis,the dermis mainly distributes in the hair follicles,glands and vessels,and the subcutaneous tissues mainly gather at the side of the blood vessels.Compared with the blank group,the 5-HT positive markers in the model group increased significantly;Compared with model group,moxibustion+model group,electro acupuncture+model group,scraping+model group,5-HT positive markers were significantly reduced;There was little difference between the intervention groups,and the positive markers of 5-HT were significantly more than those of the blank group.Compared with the blank group,the number of 5-HT positive cells marked by 10 times the microscope was significantly increased(*P<0.05).Compared with the model group,the intervention group of the three groups decreased significantly(P<0.05),and there was no significant difference between the three groups.NPY is mainly distributed in the dermis and subcutaneous tissue of the local skin tissue in the acupoint area,distributed in the dermis around the hair follicles or vessels,and in the subcutaneous tissue around the blood vessels.Compared with blank group,the expression of NPY positive fibers in model group decreased significantly;Compared with the model group,the NPY positive expression fibers in moxibustion+model group,EA+model group,scraping+model group increased significantly;There was little difference between the intervention groups,and the NPY positive fibers were significantly less than those in the blank group.Compared with the blank group,the number of NPY positive fibers in the model group and the three groups were significantly decreased(*P<0.05).Compared with the model group,the intervention group increased significantly(P<0.05),and there was no significant difference between the three groups.In the local skin tissue of the acupoint area,CGRP mainly showed a string of beads,which showed a free state near the epidermis of the superficial dermis,and the deep and subcutaneous tissue of the dermis were distributed around the blood vessels.Compared with blank group,the positive expression of CGRP in model group increased significantly;Compared with the model group,the CGRP positive fibers in the moxibustion+model group,the electroacupuncture+model group and the scraping+model group decreased significantly,and the difference between the intervention groups was not significant,and the CGRP positive fibers were significantly less than those in the blank group.Compared with the model group,the CGRP positive fibers in the moxibustion+model group,the electroacupuncture+model group and the scraping+model group decreased significantly,and the difference between the intervention groups was not significant,and the CGRP positive fibers were significantly less than those in the blank group.Conclusions:1.There are similarities and differences in response to different body stimulation methods:Electroacupuncture and moxibustion can affect the expression of 5-HT and HA in the local skin tissue of the acupoints.5-HT and HA may be the common material basis for the response of electro acupuncture and moxibustion stimulation to the acupoints;Compared with moxibustion,electroacupuncture can affect the expression of SP and CGRP in the local skin tissue of the acupoint area.SP and CGRP may be the specific material basis for the response of electroacupuncture stimulation to the acupoint area.2.The behavior experiment proved that moxibustion,electroacupuncture and scraping can improve the pain threshold of CCI model rats effectively,which shows that moxibustion,electroacupuncture and scraping have some therapeutic effect on sciatica;Under the pathological conditions,the expression of related chemicals 5-HT,NPY and CGRP in the local skin tissue of the acupoint area changed significantly.The electroacupuncture,moxibustion and scraping can make it reverse and return to the normal level.5-HT,NPY and CGRP may be the common material basis for the treatment of sciatic neuralgia by electroacupuncture,moxibustion and scraping.
