Alpinia officiniarum Hance belongs to the genusus of Alpinia in Zingiberacea family.It tastes pungent,hot and goes into spleen,stomach.It has many effects such as warming stomach,preventing vomiting,cold and pain.It's mainly used in the clinical course of treatment of pain in abdominal,stomach,cold,vomit etc.Modern clinical treated stomach pain,gastritis,ulcers and stomach cancer and other diseases.In recent years,domestic and overseas studies have shown that it had significantly inhibition of tumor.And the effective part is ethyl acetate extraction which is from its alcohol extraction.24 compounds were isolated by silica gel,ODS column chromatography,Sephadex LH-20 etc.column chromatography and HPLC modern extraction methods and technology.Their structures have been elucidated by combining the NMR spectroscopy(such.as 1H-NMR,13C-NMR,1H-1H COSY,HSQC and HMBC etc,1D and 2D-NMR spectroscopy testing techniques)with ESI-MS and HR-ESI-MS as follow.There are 5 flavonoids:Galangin(1),Galangin 3,5,7-trimethylether(2),Kaempferol(3),Quercetin(4),pinocembrin(5).And 4 diarylheptanoids as follow:(4E)-7-(4"-hydroxy-3"-methoxyphenyl)-phenyl-1-phenylhept-4-en-3-one(6),1,7-diphenyl-4E-en-3-heptanone(7),2,5-Furan-l-(4"-hydroxy-3"-methoxyphenyl)-phenyl-7-phenylheptane(8),yakuchinone A(9).5 glycoside:ethyl-?-D-glucopyranoside(10),Uridine(11),2,6-dimethoxy-4-hydroxyphenol-1-O-glucopy-ranoside(12),Staphylionoside D(13),3,5-dimethoxy-4-hydroxy benzoic acid-7-O-?-D-pyranglucose(14).6 phenolic acid compounds:p-Hydroxy benzaldehyde(15),Vanillin(16),3-methoxy-4-methylbenzaldehyde(17),4-(4'-hydroxy-3'-methoxyphenyl)-2-Butanone(18),Protoc-atechuic aldehyde(19),Protocatechuic acid(20).Compounds 10?11?12?14?15?16?17 were first isolated from Alpinia genus.Compounds 9?13?18?19?20 were first isolated from the plant.MTT assay was used to screen anti-tumor activity of each compound.Compounds were actioned in vitro-derived tumor cells:HepG-2 cells,BGC-823 cells,SGC-7901 cells,MGC-803 cells and MCF-7cells,respectively.The tests showed that IC50 value of compound 1 on HepG-2 cells was 29.98 ?M,IC50 values of compound 5 and 9 on BGC-823 cells were 22.81 and 23.08 ?M,IC50 value of compound 8 on SGC-7901 was 21.43 ?M.IC50 value of compound 9 on SGC-7901 cells was 26.90 ?M.IC50 value of compound 17 on MGC-803 cells was 66.54 ?M.IC50 value of compound 5 on MCF-7 cells was 21.43 p,M.This paper has systematic and depth studied on the chemical composition and combined the test of the anti-tumor activity of monomer compounds.To develop the anti-tumor drugs which is urgent on clinical provides important scientific basis.On this basis for the further development and use of the rich resources on Alpinia officiniarum Hance provides the possibility.
