| Objective:This paper aims at providing scientific data for the preclinical study of a traditional Kazakh herbal medicine Qufeng Babu paste by performing investigations into its preparation technology,quantification and anti-inflammatory and analgesic effects.Methods:(1)By using orthogonal experimental design,with the comprehensive sense,initial adhesion and cohesion as the evaluation index,the choice of materials the dosage of paana,glycerin and azone ratio,AI(OH)3 dosage of 3 factors,to optimize the preparation process of kazakh medicine Qufeng plaster;(2)the application of high performance liquid chromatography detection of kazakh medicine Qufeng Babu paste the main effective component of harmine,harmaline to hydrogen content and berberine,provide methodology reference for quality control;(3)with reference to"the national standard of chemical reagent(potassium hydroxide)GB/T 2306-2008 Sahar",established quality standards;(4)animal experiments were performed using the mouse test,the implementation of the writhing test,hot method,the anti-inflammatory experiments,carrageenan induced inflammation and other experiments to verify the anti-inflammatory and analgesic effects of dispelling wind,Babu ha medicine ointment.(5)the statistical treatment,the experimental data collection,analysis of the orthogonal experiment results using range analysis,variance,the animal experimental results two two comparison using t test,the test level 0.05.Results:(1)the study by range analysis results show that all the factors affecting the size of the order of A(paana)>C(glycerol and azone ratio)>B(AI(OH)3),optimization of process conditions is A1B3C2.(2)The research shows Sahar ash detection is 83.42%,the limit value of 90%;the water for an average of 12.07%,the threshold is 15%standard 18.05%;hydroxides;carbonate standard is 29.02%.(3)Harmine、harmaline and berberine calibration curve was linear over the range of13.6381.788μg·ml-l(r=0.9999,n=6)、7.4044.40μg·ml-l(r=0.9999,n=6)and13.5581.30μg·ml-l(r=0.9997,n=6),The recovery rates were 100.28%、99.59%and100.35%,RSD were 1.61%、0.97%and 1.03%.Concentration and peak area showed a good linear relationship.(4)Hazakh medicine qufeng poultice group compared with the control group,three dose group can significantly reduce the number of writhing in mice,especially in the high dose group was the most obvious(p<0.01);pain threshold of three doses induced by hot plate in mice and significantly inhibit the paw licking time after drug,3h,4h of high dose group was significantly prolonged(P<0.05);more significant anti-inflammatory effect of three dose(P<0.05);after administration of 3h,4h Hazakh medicine qufeng poultice paste of high dose group and low dose group of anti-inflammatory effect more obvious(P<0.05).Conclusion:(1)HPLC method for the determination of the experimental method of harmaline,harmine,berberine ha medicine in Babu ointment reliable,stable,and can be used for the quality identification of kazakh medicine Qufeng Babu plaster.(2)Sahar to establish a quality standard for calibrating standard ash,moisture,and carbonate and other inorganic matter;(3)Animal experimental results showed that Qufeng Babu plaster has significant.anti-inflammatory,analgesic effect on the experimental animals. |
