Pain is the body tissue damage due to internal and external environment of a kind of physiological and pathological reaction,handling to damage to embody the defensive protection mechanism of the body of pain,acute pain can cause physiological function disorder,even more serious can induce shock and even death,so the development of novel analgesic active ingredient and research become a hot research topic in recent years.As a traditional Chinese medicine with a long history,Valeriana jatamansi jones has rich ingredients,clear efficacy,small toxic and side effects,and no addiction.It has a wide range of pharmacological effects and has a high development value.However,there are few reports on the analgesic effect of Valeriana jatamansi fragrance.In this paper,the anti-inflammatory and analgesic effects of Valeriana jatamansi jones were studied,the analgesic effects of Valeriana jatamansi jones from different habitat were compared,and a HPLC method for the determination of Valeriana jatamansi jones's aurantiin was established.The following results were obtained:1.Effect of Valeriana jatamans jones extract from auricle swelling induced by xylene in mice.The effects of water extraction,water extraction(ultrasound),alcohol extraction and alcohol extraction(ultrasound)on auricle swelling induced by xylene in mice were investigated by using dexamethasone as positive control.The results showed that among the four extraction methods,alcohol extraction(ultrasound)method had the best anti-inflammatory effect,and the inhibition rate of swelling was56.94%,17.33±7.15,with significant difference(P<0.01).The three methods of water extraction,alcohol extraction and water extraction(ultrasound)showed that the swelling inhibition rates were 17.08%,39.41%and 46.92%,respectively.The swelling inhibition rates were all lower than 50%,and the swelling inhibition rates were 39.59±14.53,28.64±11.71 and 28.10±14.82,respectively,with no significant difference.2.Non-steroidal anti-inflammatory drug etopoxil was selected as the positive control to investigate the anti-inflammatory effect of alcohol extracted(ultrasonic)from Valeriana jatamans jones flavor in different dose groups.The swelling inhibition rates were 27.9%and 63.13%,respectively.The high dose group had the best anti-inflammatory effect.3.The effects of two extracts from Valeriana jatamans jones perfume(ultrasonic)and alcohol extract(ultrasonic)on mice writhing induced by glacial acetic acid were investigated.The results showed that the twist frequency of the low-dose and medium-dose groups were 19.70±4.99 and 16.08±2.91,respectively,and the difference was not significant.The twist frequency of the high-dose group was 13.25±5.97,and the difference was significant,with the analgesic rate was up to 41.32%.The twist frequency of alcohol extraction(ultrasound)in the low-dose group was17.33±4.97,with no significant difference.The median dose group was 14.78±4.71,with a significant difference,and the analgesic percentage was 34.54%.The writhing frequency of the high-dose group was 10.42±3.09,with a significant difference,and the analgesia rate was up to 53.85%.In summary,the twist frequency and analgesic percentage of the low,medium and high dose group were higher than those of the water extract group.The results showed that alcohol extraction(ultrasound)was the best extraction method.The analgesic and anti-inflammatory effects of the extracts obtained by the two methods were similar,suggesting that there was a certain correlation between the content and the analgesic effects of the anti-inflammatory active components.4.The analgesic effects of alcohol extract(ultrasonic)from six arachnids were investigated.The results showed that there was no significant difference in the number of body twisting and the percentage of analgesia was lower than 30%.In the medium dose group,yunnan and guizhou showed the best effect,with the twist frequency of 11.87±9.51 and 13.67±7.33,respectively,and the difference was extremely significant,with the analgesic rate up to 46.77%and 38.70%.In the other medium dose groups,there was no significant difference in the twist frequency,and the analgesic percentage was all lower than 30%.In the high-dose group,except for guangxi,the difference was significant in other places of origin(P<0.01),indicating that the high-dose group extracted by alcohol extraction(ultrasound)method from Valeriana jatamansi jones of all places of origin had certain analgesic effect.5.An HPLC method was established for the determination of arachnin.The linear equation was y=22.08x-61.623,R~2>0.9998.The quantitative limit and detection limited were 8.416?g/mL and 21.482?g/mL,respectively,and the RSD of precision,reproducibility and stability were 0.24%,0.277%and 0.79%,respectively.6.The content of hesperidin was positively correlated with the intensity of its anti-inflammatory and analgesic effects,suggesting that the anti-inflammatory and analgesic effects were caused by the peripheral anti-inflammatory effects of hesperidin.
