Polygala telephioides herba belongs to Polygala telephioides Willd.The whole dry grass has been commonly used in the treatment of scatter stasis hemostasis,phlegm cough,detoxicate detumescence.Currently,the basic research related to Polygala telephioides herba is still very limited.Due to lack of reference substance,its quality standard can't been improved,subsequent promotion and application of the medicine is limited severely.Polygala telephioides herba is included in the“Hebei medicinal standard”?2016 edition?.The purpose of this paper is to study the extraction method of Polygala telephioides herba and pharmacological experiment,establish a improved quality standard and lay a material foundation for controlling quality of Polygala telephioides herba.Telephenone B was isolated and purified by column chromatography technology from Polygala telephioides herba.The structure of Telephenone B was identified by1H-NMR and 13C-NMR.The purity of Telephenone B?Self made?by high performance liquid chromatography with evaporative light scattering detector?HPLC-ELSD?method was 99.2%.Using it to pharmacology experiment,the results shown that Telephenone B has significant inhibitory effect in morphine withdrawal symptoms in mice.The powder characteristics of Polygala telephioides herba were studied,and the main microscopic characteristics of Polygala telephioides herba were described.Thin layer chromatography?TLC?method was established for the identification of Polygala telephioides herba.Medicinal powder was ultrasoniced in ethanol,and the filtrate was concentrated.By treatment of polyamide column,30%ethanol elution liquid was concentrated and pointed to silica gel G thin layer plate,developing system chloroform-methanol-water?8:2:0.2?was applied.Then it was sprayed with 10%sulfuric acid in ethanol,and heated to the spot color clear in the 105?.The same color spots in the TLC graphs of sample existed at the corresponding position compared with the controls.According to the inspection items of“China pharmacopoeia”four general principles?2015 edition?,moisture content,ash content?total ash content and acid-insoluble ash content?and alcohol-soluble extraction of Polygala telephioides herba were determined.Moisture content is not more than 9.0%?general principles of the 0832,second method?.Total ash content is not more than 6.0%?general principles of the 2302?.Acid-insoluble ash content is not more than 1.5%?general principles of the 2302?.The content of alcohol-soluble extraction is not less than 20.0%by hot-dip method?general principles of the 2201?.The HPLC method and QAMS method were combined to establish the quantitative analysis method of mangiferin and Telephenone B for the first time.The column was Agilent HC-C18?4.6×250mm,5?m?;the mobile phase were acetonitrile-0.05moL·L-1sodium dihydrogen phosphate solution?1%phosphoric acid solution was adjusted to pH2.5??15:85?;the flow rate was 1.0mL·min-1;the detection wavelength was 206nm;the column temperature was 25?.Mangiferin and Telephenone B have good linear relationships respectively in 20140?g·mL-1.Experiments of the relative correction factor was fmangiferin/Telephenone B=0.662,and the relative retention value was rmangiferin/Telephenone B=1.660.The improved quality standard can be used to evaluate the inner quality of Polygala telephioides herba and guarantee its quality.
