Study On Quality Standard Of Herba Aleuritopteris

Herba Aleuritopteris has been used commonly in the treatment of abnormal menstruation and hemoptysis of pulmonary tuberculosis.Herba Aleuritopteris is effective and resource-rich,but the basic research related to Herba Aleuritopteris is still very limited,so that application of the medicine is limited.Herba Aleuritopteris is included in the“Chinese Pharmacopoeia”?2015 edition?.This paper aims to improve its quality standard and provide a technical foundation for the full and effective quality control of the medicine and preparation.In this paper,standard was isolated and purified by various chromatographic methods,such as silica gel and ODS for the first time.According to the physico-chemical properties,spectral data and the comparision with the literature data,the structure of standard was identified.Also,the standard was isolated from Herba Aleuritopteris for the first time.The purity of standard was 99.52%determined by thin layer chromatography and high performance liquid chromatography.Moisture content,ash content?total ash and acid-insoluble ash content?and alcohol-soluble extraction of medicine were determinated referring to the Appendix of“Chinese Pharmacopoeia”?2010 edition?.The content of alcohol-soluble extraction is not less than 6.5%by hot-dip method?Appendix XA?.Moisture content is not more than13.0%?first method in Appendix IX H?.Total ash content is not more than 12.0%?Appendix IX K?.Acid insoluble ash content is not more than 5.0%?Appendix IX K?.TLC method was established for the identification of Herba Aleuritopteris.Using ultrasonic extraction 30 min with petroleum ether,drawing 5?L solution on efficient silica gel G thin layer plate?100 mm×100 mm?,developing system cyclohexane-acetone?7:3?was applied.After spraying with 10%sulfuric acid in ethanol,significant purple color appeared on the sample solution with the standard at same position.For the first time,HPLC method was established for the determination of fenbeijuesu A in Herba Aleuritopteris.The mobile phase were acetonitrile and water?7:3,v/v?;the flow rate was 1.0 mL·min^-1;the detection wavelength was 202 nm;the column temperature was 40?.The linearity range was 0.04-8.0?g?r=0.9998?.The mean recovery was 100.20%?RSD=2.90%?.The limit of content was determined according to the requirements of the Pharmacopoeia.
