Objective: Eucommia ulmoides(Eucommia ulmoids Oliv.)is a dioecious tree belonging to only one species,one genus(Eucommia)and one family(Eucommiaceae),which is sweet,warm and non-toxic.It is known as Mumian,which has 2000 years of history in China,is a traditional Chinese herbal medicine.Eucommia belongs to the rare and endangered species in China,the bark is often used for medicine,it grows more slowly,but it will be die when peeling the trees,which has brought a great inconvenience to protect traditional Chinese herbal medicine resources.There are a lot of leaves and it is very easy to pick them,so in recent years,many domestic and foreign scholars are contribute to researching it.The bark and leaves after drying have the similar components and pharmacological effect.In the year of 2005,it has been listed “Chinese Pharmacopoeia”,it could nourish liver and kidney,strengthen bones and muscles,and sooth the fetus.Excessive intake of alcohol have a serious impact on physical and mental health,it can increase free radicals and change the biological activity of the enzyme in vivo.It also can damage the central nervous system and inhibit the brain's function.Now scholars focus on the ethanol-induced DNA damage,but the extraction of alcohol-induced brain damage protection study is less,therefore,the research object is Eucommia ulmoides leaves in this experiment to research the protective of effect on alcohol-induced brain tissue in mice.Methods: in this study,we used kunming mice as the experimental subjects,which divided into five groups: control group,positive control group,high and low dose group and model group.High and low dose group and positive control group administered continuously for three days.On the third day administered half an hour after intragastric given the ethanol(6g/kg),while the control group and the model group were given the appropriate volume of 0.5%CMC-Na solution.They were dislocated executed after four hours.By single cell gel electrophoresis(comet assay)to screen the leaves 95% alcohol extract and 50% alcohol extract and water extract of protection due to ethanol cause mice brain regions of DNA damage.By enzyme-linked immune method and colorimetric method to detect the crude extract of induced by ethanol in mice,the cerebral cortex,cerebellum,plasma and liver tissue physiological indexes related to oxidative damage,such as NO(nitric oxide synthase),MDA(malondialdehyde)and 8-OHDG(8-hydroxy deoxyguanosine)content,T-SOD(total superoxide dismutase),GSH-PX(glutathione peroxidase)activity,NQO1(quinone oxidoreductase1)level and ChAT(cholineacetyl transferase).According to the literature,it contains more polyphenols antioxidant ingredients,so by the Folin ciocalteu colorimetric method to detect the total polyphenols content in 50% alcohol extract.Results: in the comet assay stage,with the relevant professional software and found that the effect of different solvent extracts antioxidant are also different,the best effect is the 50% ethanol extract.After giving the 50% ethanol extract to detect the biochemical indicators,such as MDA,NO,NQO1,T-SOD,GSH-PX and so on.The results have showed that after administration,the capacity of antioxidant have the same trend in mouse brain regions,plasma and liver tissue,but have a clear dose-response relationship.The 50% alcohol extract could significantly reduce the plasma and liver tissue and brain tissue NO and MDA content and also effectively increase the activity of T-SOD,GSH-PX and NQO1,indicating that the alcohol extract can improve the tissue antioxidant capacity to inhibit free radical damage to cells,tissues and reduce the formation of oxidation products to achieve the effect of DNA protection.This experiment is mainly using the vitamin E as the positive control.We have found that the alcohol extract of the bark of Eucommia ulmoides and vitamin E can increase the body resistance and removal of free radicals in the body.By the Folin ciocalteu colorimetric method,which determine about 61.6% total polyphenols in it,indicating that the protection of DNA and antioxidant effect in total polyphenolic components play an important role.Conclusion: acute ethanol administration can produce a strong neurotoxicity and accelerate the free base generation,so that the body generates less antioxidants.Through a variety of kits testing have found that ethanol can affect some physiological indexes in human body,making certain oxidation indexes such as NO,MDA content increased,antioxidant indexes such as T-SOD,GSH-PX and NQO1 activity decreased.By the experiment,we have found that it can eliminate the free radicals and reduce DNA damage,and then enhance the activity of the T-SOD,GSH-PX and NQO1 and effectively reduce the peroxidation to play a role in neuroprotection.
