Objective: 1.To observe the effects of Eucommia ulmoides leaves extract and Gardenia extract on the changes of body weight,blood lipid,inflammatory factors and visceral pathology in mice induced by monosodium sodium Glutamate.2.To observe the effects of eucommia ulmoides extract and gardenia extract on glucose and lipid metabolism in monosodium Glutamate-induced obese mice.3.To study the effects of eucommia ulmoides extract and gardenia extract on bile acid-related transporter in monosodium Glutamateinduced obese mice.Methods: 1.Subcutaneous injection of sodium Glutamate(4 g/kg/day)intervened growth in mice,and normal control group mice were subcutaneous injected saline(10 ?L/g/day).After 4 weeks,mice were divided into 9 groups: normal control group(Normal),model group(Model),positive control group(RES),high dose group of Eucommia ulmoides leaves extract(EEH),low dose group of Eucommia ulmoides leaves extract(EEL),high dose group of Gardenia extract(GEH),low dose group of Gardenia extract(GEL),high dose group combination(EEH+GEH),and low dose group of combination(EEL+GEL),10 mice in each group.The glucose tolerance and insulin tolerance,body weight,body length and waist circumference were measured in eighth weeks.And then,the serum lipids,inflammatory factors and liver function were measured.HE staining was used to observe the morphological changes of fat in liver,small intestine and epididymis.2.Real time quantitative PCR assay was used to detect the expression of m RNA in glucose and lipid metabolism related genes in obese mice.3.Real time quantitative PCR assay was used to detect the changes of m RNA expression in the bile acid transporter gene of each group,and the changes of bile acid transporter protein were detected by Western Blot method.Results: 1.After 8 weeks of treatment,compared with the normal control group,the model group mice of body weight,waist circumference,Lee's index and epididymal fat were increased,the difference was statistically significant(P<0.05);and TC,TG,LDL-C,IL-6,IL-1,ADP,TNF-?,ALT and AST increased significantly,and HDL-C decreased in different extent,and at the same time the leptin and insulin resistance.The body weight,waist circumference,blood lipid,inflammatory factors and insulin resistance were improved in different groups.Among them,the high dose group was the most obvious(P<0.05).HE staining showed that there were a lot of lipid droplets in hepatocytes of the model group,and liver cells arranged in disorder,and the necrotic changes of the central lobe,and hepatic lobules.HE staining showed that compared with the control group,model group of the intestinal glands of different lengths,gland malalignment,intestinal mucus increased slightly.In the model group,HE staining showed the number of epididymal fat cells increased in different degree,and the number of cells in the same visual field decreased significantly,while the administration group could improve the symptoms in different degree(P<0.05).2.The expression of lipid metabolism genes in liver and small intestine detected by real time fluorescent quantitative PCR in m RNA showed that each group in the liver and intestine can upregulate PPAR?,PPAR? and GLUT,and decrease SCD1,FASN,SREBP-1c,PPARgc1?,PEPCK and G-6-P.3.Bile acid transporter associated with real-time fluorescence quantitative PCR method for detection of heart,liver,kidney and small intestine in the expression of m RNA showed that the expression of TGR5 of m RNA can be up regulated in different degree,but in the epididymal fat is not obvious;the different degrees of increase the expression of OST?,OST?,ABCG5 and ABCG8 in liver and intestine;the expression of CYP7A1,Slco1b2,Slco1a1,MRP2 and MRP3 increased in the liver;different degree of reduction of FXR and SHP in the intestine and liver m RNA expression;decreased the expression of FGF15 and FGFR4 m RNA in the small intestine.Western blot analysis showed that the protein expression of bile-related transporters in the heart,liver,kidney and small intestine of TGR5,NTCP,ABCG5 and ABCG8 were up-regulated in different degrees,and the bile acid synthesis gene Protein expression were up-regulated;and down-regulated the expression of protein of FXR,SHP,BSEP,AKT,m TOR,NK-?B and i NOS.Conclusion: In view of the above,the administration of Eucommia ulmoides extract and Gardenia extract improved the body size,body weight,the pathological changes of liver,intestinal and adipocyte,blood lipid levels,inflammatory factors and liver function in obese mice induced by monosodium sodium Glutamate in a dose-dependent manner.The mechanism may be through the impact of glucose and lipid metabolism related gene expression,while regulating the bile acid-related transporter and inflammation-related gene expression and protein expression.
