Objective: To study the effects of Liensinine on T24 cells cycle and preliminary study of its mechanism.Methods: Treat bladder cancer cell line T24 with Liensinine solution,the concentration were(1.562,3.125,6.250,12.500,25.000 ?g/?L)respectively.CCK-8 kit and the colony formation test was done to detect cell viabilities after treatments and then inhibition rates were calculated.Flow cytometry was used to detect the effects of Liensinine on cell cycle of T24 cells after the treatments.Western blot was used to detect p27 expression.Results: CCK-8 assay indicated that With the increasing concentration of the drug and prolonged incubation time,Liensinine inhibited the T24 cell proliferation significantly.After treatment of 48 h,the proliferation inhibition rates were(8.79±0.07)%?(16.11±0.05)%?(21.05±0.07)%?(27.79±0.05)%?(41.46±0.04)%.The difference was statistically significant(p<0.05).Colony formation test also showed that Liensinine could inhibit the proliferation of T24 cells.The proliferation inhibition rates were(20.34±0.32)%?(57.95±0.19)%?(89.98 ± 0.45)% ?(94.22 ± 0.02)% ?(95.16 ± 0.01)%,The difference was statistically significant(p<0.05).Flow cytometry indicated that Liensinine could significantly inhibit the proliferation of T24 cells and arrest the T24 cells at the S phase.The rates of the S phase in different doses groups were(34.03±0.85)%?(35.49±2.41)%?(37.89±0.55)%?(41.29±1.18)%?(54.48±1.93)%?(58.78±0.38)%.Western blot showed that the activation of p27 in dosing groups had improved significantly.Conclusions: Liensinine could inhibit the proliferation of T24 cells and arrest the T24 cells at S phase,the mechanism may be the enhanced expression of p27 and its activation.
