The Effects Of Lactated Ringer's Solution And Acetated Ringer's Solution Resuscitation Of Hemorrhagic Shock In Rats On Its Inflammatory Cytokines

Objective:To compare the effect of intravenous resuscitation with acetated Ringer(AR)solution and lactated Ringer solution(LR)on TNF-?,IL-6 and IL-10 during hemorrhagic shock(HS)resuscitation in rats.Methods: Twenty-four SD rats were equally randomized into 4 groups: No Resuscitation(NR,n=8);Acetated Ringer solution resuscitation(AR,n=8);Lactated Ringer solution resuscitation(LR,n=8).Model of HS were made in the latter two groups and subjected to different types of fluid resuscitation respectively.Peripheral blood of rats in latter two groups were collected for five time node: before shock(T0),after 60 minutes of keeping HS(T1),after 30 minutes of resuscitation(T2),after 60 minutes of resuscitation(T3).NR group at the corresponding time collected blood,and liver and lung tissue were taken at end of the experiment.ELISA method was used to measure TNF-?,IL-6 and IL-10 content in plasma and liver,lung tissue.Results: The peripheral blood IL-10 content of each group at T0,T1 and T2 time point had no significant difference(P> 0.05).At T3 time point,the IL-10 content of NR group was significantly higher than LR group(P <0.05);no significant difference is between LR group and AR group(P>0.05).The TNF-?,IL-6 content of each group at T0,T1 time point had no significant difference(P> 0.05).At T2 time point,TNF-?,IL-6content of NR group was higher than both LR and AR groups(P <0.05);no significant difference is between LR group and AR group(P>0.05).At T3 time point,TNF-?,IL-6content NR group was significantly higher than LR group(P <0.05)and LR group was also higher than AR group(P < 0.05)..The TNF-?,IL-6 content of liver and lung tissue,NR group were significantly higher than both LR and AR groups(P < 0.05)and LR group was also higher than AR group(P < 0.05).The IL-10 content of liver tissue,NR group was significantly higher than both LR group and ARgroup(P <0.05);no significant difference is between LR group and AR group(P>0.05).The IL-10 content of lung tissue,NR group was significantly higher than LR group(P < 0.05);no significant difference is between NR group and AR group(P>0.05);no significant difference is between LR group and AR group(P>0.05).In liver and lung tissue,the score of NR group was significantly higher than LR group and AR group(P <0.05).Lung tissue of lactated Ringer resuscitation score is significantly higher than AR resuscitation group(P <0.05);Liver tissue of lactated Ringer resuscitation score is higher than sodium acetate Ringer resuscitation group(p> 0.05).Conclusion:In the process of resuscitation,AR can better suppress the release of TNF-?,IL-6 in plasma.AR can better suppress the release of TNF-?,IL-6 in liver and lung tissue.The AR can better protect liver and lung tissue.The AR was superior to the LR in improving the release of inflammatory mediators for resuscitation in rats with HS.
