ObjectiveUsing an animal model of vascular hyperpermeability in ovariectomized rat to observe the effects of Soybean Isoflavones Genistein(Gen),Biochanin A(Bio)on endometrial exudation,the changes of endometrial structure and the expression of vascular endothelial growth factor(VEGF)induced by exogenous 17?-estradiol and to elucidate the related mechanisms.MethodsFifty-four SD rats were randomly divided into 9 groups,each group had 6 rats:Sham,Ovx,Ovx+Est 1.0 mg·kg-1 body weight(Ovx+Est),Ovx+Est 1.0 mg-kg-1 body weight+Gen1.0 mg-kg-1 body weight(Ovx+Est+Gen1.0),Ovx+Est1.0 mg·kg-1 body weight+Gen10.0 mg-kg-1 body weight(Ovx+Est+Gen 10.0),Ovx+Est 1.0 mg·kg-1 body weight+Bio5.0 mg·kg-1 body weight(Ovx+Est+Bio5.0),Ovvx+Est1.0 mg·kg-1 body weight+Bio10.0 mg·kg-1 body weight(Ovx+Est+Bio10.0),Ovx+Est1.0 mg·kg-1 body weight+Tam30.0 mg·kg-1 body weight(Ovx+Est+Tam30.0),Ovx+Est1.0 mg·kg-1 body weight+Sut40.0 mg·kg-1 body weight(Ovx+Est+Sut40.0),After administration of different estrogens for 21 days,we did the following detections:1.observed endometrial morphological changes;2.counted the eosinophilic granulocyte;3.surveyed the VEGF expression of every group using Western blot and Immunohistochemical technique.Results1.Changes of endometrial morphologyAfter the ovary removal,we found the rat uterus was atrophic,endometrium and myometrium were thin,the myocyte diameter was decreased.After administration of exogenous 17?-estradiol,the endometrial hyperplasia appeared,myometrium was thick,the myocyte diameter was increased(P<0.01).To compare with Ovx+Est group,endometrium and myometrium were thin in Ovx+Est+Gen1.O,Ovx+Est+Bio5.0,Ovx+Est+'Tam30.0 and Ovx+Est+Sut40.0 groups,and the myocyte diameter was decreased(P<0.01),but in both of'Ovx+Est+Gen10.0 and Ovx+Est+Bio10.0 groups,endometrium and myometrium were thick,the myocyte diameter was increased,there was no obvious difference among Ovx+Est+Gen 10.0.Ovx+Est+Bio10.0 and Ovx+Est groups.2.The exudation of eosinophilsAfter the ovary removal,we found the exudation of eosinophils in endometrium reduced.After administration of exogenous 17(3-estradiol,the exudation of eosinophils increased.To compare with Ovx+Est group,the exudation of eosinophils in endometrium reduced in Ovx+Est+Gen1.0,Ovx+Est+Bio5.0,Ovx+Est+Tam30.0 and Ovx+Est+Sut40.0 groups.There was no obvious difference among Ovx+Est+Gen10.0,Ovx+Est+Bio10.0 and Ovx+Est groups.3.The observation of VEGF expression in uterine tissueThe endometrial expression of VEGF was localized in the glandular,endometrial epithelium and mesenchymal cells;VEGF was expressed at different level in nine groups.To compare sham group with Ovx group,the expression of VEGF was down-regulated in Ovx group.To compare with Ovx+Est group,Ovx+Est+Gen1.0,Ovx+Est+Bio5.0,Ovx+Est+Tam30,0 and Ovx+Est+Sut40.0 groups,the expressions of VEGF were down-regulated.There was no obvious difference among Ovx+Est+Gen10.0,Ovx+Est+Bio10.0 and Ovx+Est groups.ConclusionsDifferent doses of soy isoflavones genistein and biochanin A performance different role.Low doses can inhibit the proliferation,exudation and VEGF expression in the endometrium induced by exogenous 17?-estradiol in ovariectomized rats,has estrogen antagonistic role.High doses have no inhibitory effect on 17?-estradiol.
