ObjectiveTo study the effect of Huang Qi San on lipodystrophy-induced insulin resistance mice and its mechanism,and provide a foundation to clarify its mechanism on improving insulin sensiti-vity.MethodThe lipodystrophy model(aP2-SREBFlc)transgenic mice were used to observe the effect of HQS on glucolipid metabolism related parameters.After adimistration,to determine the content of plasma glucose and lipid with conventional biochemical detection methods.To carry on the oral glucose tolerance test(OGTT)and insulin tolerance test(ITT)and observe the change of glucose tolerance and insulin resistance in mice,using the method of tissue homogenate to detect lipid content changes within the relevant organ,Elisa method was used to determine the change of fasting plasma leptin and insulin levels.Reverse transcription polymerase chain reaction(RT-PCR)technique was used to detect the expression quantity of glucolipid metabolism related genes mRNA expression in liver and adipose tissue.ResultsThe lipodystrophy model ap2-SREBP-lc mice were observed disordered differention of adipose tissue.The size of white adipose tissue was markedly decreased.Plasma leptin was reduced,and the levels of plasma glucose and insulin were increased.Plasma triglyceride and total cholesterol level were elevated.The transgenic mice were glucolipid metabolic disorders,obvious insulin resistance and fatty liver disease.HQS neither improve the level of plasma glucose and insulin,nor glucose tolerance and insulin tolerance.HQS combined with leptin reduced the plasma glucose in mice,and HQS combined with leptin increased insulin sensitivity of ap2-SREBP-lc mice in ITT;According to OGTT,HQS combined with leptin improved glucose tolerance in mice.The above information prompts the HQS powder to improve insulin sensitivity may rely on leptin play a role.With the results of Liver pathology detection and lipid content determination,HQS combined with leptin reduced hepatic lipid ectopic deposits.Reverse transcription polymerase chain reaction(RT-PCR)detection results showed that HQS combined with leptin increased PPARy,reduced SREBP-lc in adipose tissue;FAS,PPAR?,SCD1 and G6PC expression decreasing in the liver,while increased the expression of PPARa,CPT1a,HSL.ConclusionHQS combined with leptin reduced plasma glucose and lipid in lipodystrophy-induced insulin resistance mice,relieved ectopic lipid deposition and glyconeogenesis,and enhanced insulin sensitivity,improve glucolipid metabolism.The internal mechanism of HQS may be associated with leptin.
