Objective:To explore the curable effect of PND on mice with immune mediated apalstic anemia,to study AA with MAPK/ERK signal transduction pathway relevant.Methods:Balb/c mice were irradiated 5.0 Gy(1Gy/min 5min)with 60Co-? rays.4 hours later,the mice were injected lymphocytes mixed thymus,lymph nodes of the DBA/2 mice(the concentration was 5 × 106/ml)0.2 ml through Balb/c mice vena caudalis.60 Balb/c mice were randomly divided into normal control group?modeling group?PND low middle and high dose treatment groups and the androgen group.Observe the peripheral blood leukocytes hemoglobin and platelets of tail vein blood of mice on the 14th day;Killing the mice on the 14th day,take bone marrow biopsy of both sides of mouse femurs and observe the bone marrow smears by HE staining method.Observed MEK1/2,P-MEK1/2,ERK1/2,P-ERK1/2 of each group of mouse bone marrow protein by immuno histochemical and Western blot experiment.Results:(1)The blood cells counts were significantly decreased in peripheral blood of aplastic anemia group compared to the normal control group,and bone marrow hematopoietic cells were decreased in marrow of aplastic anemia group compared to the normal control group,indicating that mice with immune mediated apalstic anemia is successful.(2)PND could increase the peripheral blood cells of mice with immune mediated apalstic anemia.The peripheral blood leukocytes hemoglobin and platelets of PND treatment group were higher than the aplastic anemia model control group.(3)PND could promote the recovery of hematopoietic function of mice with immune mediated apalstic anemia.By the comparison of the biopsys,the marrow tissue sections of mice with immune mediated apalstic anemia appeared light staining of the hematopoietic tissue,extremly decrease of bone marrow hyperplasia and increase of adipocytes.With the dose increasing of PND,the hematopoietic tissue was gradually repaired,the hematopoietic cells were gradually increased and the adipocytes were significantly decreased.(4)PND could promote the expression of MEK1/2,P-MEK1/2,ERK1/2,P-ERK1/2,promote the recovery of hematopoietic function of mice with immune mediated apalstic anemia.Conclusion:PND can improve the peripheral blood and bone marrow of mice with immune mediated apalstic anemia.MAPK/ERK signal transduction pathway relevant protein disparate expression maybe one Pathogenesis of apalstic anemia.Up regulation MAPK/ERK signal transduction pathway relevant protein expression maybe one reason that PND could promoting bone marrow hematopoietic.
