Aplastic anemia(AA)is a disease characterized by bone marrow hematopoietic failure caused by various causes,characterized by hypoplasia of hematopoietic stem cells and reduction of whole blood cells in peripheral blood.The pathogenesis of AA is still unclear,but more and more scholars are beginning to pay attention to the important role of immune abnormalities in the development of the disease,especially the immune disorders caused by Treg and Th17 cell disorders are considered to be the key to the pathogenesis of AA.The treatment of AA in modern medicine is mainly transplantation and immunosuppressive therapy,but both of them have their own limitations and shortcomings.In the long-term discussion,TCM has a unique understanding of AA.Traditional Chinese aplastic anemia plaster was producted by Su Wei,who is the director in the Department of Hematology in Oriental Hospital of Beijing University of Chinese Medicine.The plaster has achieved good results in clinical application,also Previous studies have found it can effectively change the immune abnormal state of AA,but whether it improves the imbalance of Treg/Th17 in AA,thus achieving immunomodulatory effects,still need to explore further.Exploring the pathogenesis of AA is the basis for seeking a good treatment for the disease,the exploration of the mechanism of action of the Traditional Chinese aplastic anemia plaster can provide better data support for clinical medication and provide theoretical basis and treatment ideas for future treatment.Purposes:By establishing a good and stable animal model of AA,compare the general condition and peripheral blood picture of AA mice after drug treatment,explore the effect of Atraumatic cream on AA,and observe the expression levels of Treg and Th17 and spleen in peripheral blood of each treatment group.Morphological structure,explore the mechanism of action of the aplastic barrier.Methods:1.BALB/c mice were randomly divided into three groups,one group was given Busulfan 18 mg/kg intragastrically and IFN-y7500U intraperitoneally injected(continuously 10 days),and one group was administered with Busulfan 36 mg/kg intragastrically(continuously 5 days),a group was administered with Busulfan 36mg/kg intragastrically(continuously 7 days),comparing the quality of life and peripheral blood cell changeof each group during the administration period and 10 days after administration,also analyze bone marrow morphology Learn to judge whether the model is successful or not,and choose the better method.2.The successful AA mouse model was treated with three different treatments:Traditional Chinese aplastic anemia plaster,CsA and those two drugs combination.After 10 days of continuous intervention,three goups were observed by the quality of life and peripheral blood cell changes of AA mice.In order to analyze the efficacy of these three different treatments.By expressing Treg and Th17 cells in peripheral blood and the levels of spleen index,spleen morphological structure were observed in the model group and each treatment group,to explored the change of Treg and Th17 cells in AA,and explore whether the Traditional Chinese aplastic anemia plaster can exert immunomodulatory effects by improving the imbalance of Treg and Th17 cells.The results:1.(1)Busulfan 18mg/kg intragastrically and IFN-y7500U intraperitoneal injection(continuously 10 days)can not form an effective AA model after 10 days of intervention;(2)Busulfan 36mg/kg intragastrically(continuously 5 days)can form a stable AA model on the 5th day after the administration of the model,and can be maintained until the 10th day.(3)Busulfan 36mg/kg intragastrically(continuously 7 days)can also form AA model,but the mortality rate is high.2.In three different treatments of AA(1)Three treatments can effectively restore the general condition of the body,and there is no significant difference between the three.(2)To the peripheral blood cells of AA mice,three treatments were different effects.The WBC,RBC,HGB and PLT in the peripheral blood were recoved after the combination of the two drugs,and in the group of used the CsA alone the RBC and PLT indication was prominent.The effect of the Traditional Chinese aplastic anemia plaster on PLT was more obvious.(3)The combination of traditional Chinese and Western medicine in the treatment of AA mice is better than WBC and PLT in the treatment of CsA alone,also more obvious than only use the Traditional Chinese aplastic anemia plaster.(4)In improvement of the WBC,RBC,HGB and PLT in peripheral blood of AA mice,there is no difference between the Traditional Chinese aplastic anemia plaster and CsA.(5)The method of using Busulfan 36mg/kg intragastrically(continuously 5 days)can obtain a AA model,in which the proportion of Treg cells decreased,and Th17 cells increased.The changes of the two cells were consistent with the literature reports.(6)The effect of the combination of Traditional Chinese aplastic anemia plaster and CsA on Treg was obvious,but the effect of the two methods alone was not significant.(7)The treatment methods improved the spleen morphology of AA model mice,and the combination of the two drugs was the most obvious,followed by CsA,and the Traditional Chinese aplastic anemia plaster was the weakest.Conlusions:BALB/c mouse AA model formed by busulfan 36mg/kg/day(continuous administration for 5 days),after 10 days of drug intervention,the aplastic barrier was combined with CsA in improving the general condition of the model and peripheral blood.The use of the two is better than the two alone,and there is little difference between the two applications alone;in addition,the combination of aplastic barrier and CsA can effectively improve the imbalance of Treg and Th17 cells in AA,suggested that the combination of the two drugs can produce a therapeutic effect on AA mice by improving the imbalance of Treg and Th17.
