Objective We build this stable and reliable cold-heat combination syndrome model by experimental research,and test related serum biochemical indicators and the herb pairs'effective components' pharmacokinetic parameters which are used to characterize the herb pairs' intervention effect,and we analysis the herv pairs'impact to cold-heat combination syndrome model,so that we provide experimental basis for the usage of the treatment of combined therapy of cooling with warming in cold-heat combination syndrome.Methods We give a morning gavage on model rats of propylthiouracil(100 mg/kg),after 30 min of the gavage,put the model rats into 2? water and let them swim until model rats can't hold on(swim once every other day),and give a afternoon gavage on model rats of 100%rhizoma anemarrhenae-gypsum water decoction(4ml per rat),the ratio of rhizoma anemarrhenae-gypsum of 3:4.After 28 days of the gavage,model rats start to appears as obvious cold syndrome,and by using subcutaneous injection of 20%suspension liquid of dried yeast to cause rats get fever,10 ml/kg,we build the single fevering cold-heat combination syndrome model;we give rats subcutaneous injection of 20%suspension liquid of dried yeast every 19 hours,10 ml/kg,we build the continuous fevering cold-heat combination syndrome model.and after given radix aconiti carmichaeli?coptidis rhizome?radix aconiti carmichaeli-coptidis rhizome decoction lavage,collecting blood from rats' eyes,testing related serum biochemical indicators and the herb pairs'effective components'pharmacokinetic parameters to characterize the herb pairs' intervention effectResults Model rats after 28 days of the establishment of a cold-heat combination syndrome model by compound factors start to appears as emaciation?temperature hypothermia and a series of obvious cold syndrome,and after subcutaneous injection of dried yeast suspension liquid,the body temperature of model rats increased significantly.Compared with the blank group,model group's serum T3?T4?MDA?SOD significantly decreased,NO?IL-1?TNF-? significantly increased,and after given radix aconiti carmichaeli?coptidis rhizome?radix aconiti carmichaeli-coptidis rhizome decoction lavage respectively,medicated group rats' serum NO?MDA?IL-1?TNF-? significantly decreased,SOD significantly increased compared with model group,and had significant difference;as to NO?SOD?TNF-?,the radix aconiti carmichaeli-coptidis rhizome group'curative effect is the most significant,we can conclude that radix aconiti carmichaeli-coptidis rhizome had a positive intervention effect on rats.The three types of aconitine'drug-time curve all appear multimodal,and decline slowly in fluctuations.Berberine hydrochloride stay in model group rats for a long time,may be related to the frail of the rats after modeling,and the drug absorption is slow,so it extended the detention time.Conclusion We established a cold-heat combination syndrome model by compound factors,and we find that radix aconiti carmichaeli-coptidis rhizome has proved effective in therapy,its mechanism may be through increase the activity of SOD and reduce the content of NO?MDA,remove the body's free radicals,improve the body's antioxidant capacity,and restrain of lipid peroxidation.
