Background and objective Regulatory T cells(Treg)as a kind of immune regulation function of the mature T cell subsets,it plays a key regulatory role in the immune response through the process of inhibition on cellular immunity.Compared with other immune cells,CD4+CD25+Treg have immune features unique,mainly mediated immunologic response,induced by helper T cells through T cell receptor signaling activated(Th)1/Th2 drift,then influence inflammatory outcomes.Many studies have shown,some soluble molecules and membrane binding molecules on Treg function plays an important role in the activity,including interleukin(IL)-2,IL-4,IL-10,transforming growth factor(TGF)-?,cytotoxic T lymphocyte associated antigen(CTLA)-4 and glucocorticoid induced tumor necrosis factor receptor(GITR)etc.At the same time,forkhead/winged helix transcription factor(Foxp3)mRNA and protein specific expression in CD4+CD25+Treg,directly affect the phenotype and activity of Treg.This experiment investigated the impact of the flavonoids from Radix Tetrastigmae on serum IL-4,IL-10,TGF-? content of C57BL/6 mice with Lewis lung carcinoma and its antitumor effect.Preliminary study of the flavonoids from Radix Tetrastigmae regulate immunity function of tumor mice,reduce tumor immunosuppression and the possible mechanism of reversion of tumor immune escapeMethods The first,establishment of mouse lung tumor animal model by subcutaneous inoculation of tumor cells,were randomly divided into normal group,tumor bearing saline group,celecoxib group,flavonoids from Radix Tetrastigmae in high dose group,middle dose group of flavonoids from Radix Tetrastigmae,flavonoids from Radix Tetrastigmae in low dose group,after inoculation with 24h began intragastric administration,a once per day,0.3ml,the normal group were not gastric lavage.Second,fourteenth days later,a group of mice were killed,stripping the tumor weight and calculate the inhibitory rate of tumor inoculation.Third,collection of peripheral blood in mice,enzyme linked immunosorbent assay(ELISA)method for the determination of serum IL-10,IL-4 and TGF-?.Fourth,using SPSS Statistics 17 software package for statistical analysis of the data,the mean and the standard deviation for measurement data,among groups were compared by analysis of variance(One-way ANOVA),homogeneity of variance among groups were compared using two two minimum range method(LSD method),unequal variances among groups were two two compared with Dunnett's T3 method,any of the two group were compared using independent samples t test.Test level of a=0.05,P<0.05,said the difference was statistically significant.Results The tumor inhibition rate:clover flavonoids high dose,middle dose and low dose group,the inhibition rates were 56.3%,27.8%,6.7%,celecoxib group inhibitory rate was 53.4%.The content of serum IL-4:saline group was the highest,flavonoids from Radix Tetrastigmae in high dose group,middle dose group,low dose group,celecoxib group and normal group compared with the saline group Respectively,the content of serum IL-4 were reduced to varying degrees,every comparison was statistically significant.The content of serum IL-10:saline group was the highest,flavonoids from Radix Tetrastigmae in high dose group,middle dose group,low dose group,celecoxib group and normal group compared with the saline group Respectively,the content of serum EL-10 were reduced to varying degrees,in addition to the low dose group compared with the normal saline group had no statistical significance(p=0.239),the other had statistical significance comparison of.Serum TGF-?content:saline group was the highest,flavonoids from Radix Tetrastigmae in high dose group,middle dose group,low dose group,celecoxib group and normal group compared with the saline group Respectively,the content of serum TGF-? was decreased with different degrees,except the low dose group and the saline group showed no statistical significance(p=0.411),the other groups are significant.Conclusions The flavonoids from Radix Tetrastigmae have anti-tumor effect,and the inhibitory effect was dose dependent.The flavonoids from Radix Tetrastigmae can reduce serum IL-4/IL-10 and TGF-? in mice with Lewis lung carcinoma,and there is a certain dose dependen.The flavonoids from Radix Tetrastigmae can improve mice tumor immunosuppression.The flavonoids from Radix Tetrastigmae and celecoxib may have the same or similar approaches in anti tumor effect.
