Objective:To investigate the effects of Gypenosides on expression of mouse podocytes induced by high glucose nephrin,VEGF mRN A.Methods:conditional permanent resident mouse podocytes cultured in 33 C passage,37℃ differentiation after 14 days culture fluid replacement,containing 1%fetal bovine serum,5.5mmol/L glucose,the synchronization of cells after 24 hours,were randomly divided into:(1):groupA:normal glucose(NG,lucose 5.6mmol/L)group;(2)B group:control group(30mmol/L hypertonic mannitol);groupC:high glucose(HQ glucose 30mmol/L)group,group D:group and high dose group:HG+GPs(glucose,30mmol/L+Gypenosides,Gpsl2ug/ml),group E:group HG+GPs(glucose,30mmol/L-Gypenosides,Gps6ug/ml))groupF:HG+GPs low dose group(30mmol/L+Gypenosides glucose,Gps3ug/ml);groupG HG+10-5mol/L valsartan(HG+VAL10-5mol/L)group,groupH:HG+10-6mol/L valsartan(HG+VAL10-6mol/L)group,groupI HG+10-7mol/L valsartan(HG+VAL10-7mol/L)group,respectively for 24 hours.RT-PCR was used to detect nephrin and VEGF in mouse podocyte mRNA expression level.Results:1.after 24 h of stimulation,compared with normal glucose group,decrease the expression of nephrin mRNA in high glucose group(P<0.05 or P<0.01),VEGF mRNA expression was increased,nephrin,expression of VEGFmRNA was no significant change in the contrast group.2.compared with HG group,total saponins of Gynostemma pentaphyllum intervention can improve the expression of cultured mouse podocytes,nephrin mRAN VEGFmRNA expression,and has a certain concentration dependent(P<0.05 or P<0.01);low concentrations of Gypenosides effects were not significant(P<0.05)3.Valsartan regulating VEGFmRNA expression and gypenosides had no significant difference(P>0.05),Valsartan regulated nephrinmRNA expression significantly than Gynostemma pentaphyllum(P<0.01).Conclusion:l.High glucose can make decreased the expression of VEGF mRNA increase the expression of nephrin mRNA in cultured podocytes;2.Gynostemma total saponin and valsartan can also significantly increase the expression of nephrin mRNA induced by high glucose,overexpression of VEGF mRNA inhibited podocyte secretion,suggesting gypenosides protect renal function and partly through up-regulation of nephrin in podocyte downregulation of VEGF expression,to realize;3.There are some differences between 3 different doses of Gynostemma pentaphylla total saponin,high dose group was superior to low dose group,and the effect is similar with valsartan.
